Description: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components. This process is negatively regulated by TOR (Target of rapamycin) through phosphorylation of autophagy protein APG1. ATG16, another member of the autophagy protein family, forms a complex with the ATG5-ATG12 conjugate. This multimeric protein has been shown to be essential for autophagosome formation in both yeast and mammals and targets the ATG5-ATG12 complex to the autophagic isolation membrane during the formation of the autophagosome. Because mammalian ATG16 has seven WD-repeats in its C-terminal domain, it has been suggested that these may form a platform for further protein-protein interactions. Multiple isoforms of ATG16 are known to exist.

Description: Caspases are a family of cysteine proteases that can be divided into apoptotic and inflammatory caspase subfamilies. Unlike the apoptotic caspases, members of the inflammatory subfamily are generally not involved in cell death but are associated with the immune response to microbial pathogens. Members of this subfamily include caspase-1, -4, -5, and -12 and can activate proinflammatory cytokines such as IL-1b and IL-18. Caspase-14 is highly expressed in embryonic but not adult tissues. It is processed and activated by caspase 8 and caspase 10 in vitro, and by anti-Fas agonist antibody or TNF-related apoptosis inducing ligand in vivo. The expression and processing of this caspase may be involved in the keratinocyte terminal differentiation, which is important for the formation of the skin barrier.

Description: SPATA1, a novel cell-surface protein, is involved in shaping the sperm head during spermatogenesis. SPATA1 is localized at chromosome 1q21. At least three isoforms of SPATA1 are known to exist. The longest is preferentially expressed in hematopoietic tissues. Its extracellular domain contains a single immunoglobulin-like domain, and its intracellular segment has two immunoreceptor tyrosine-based inhibition motifs (ITIMs). It is thought to have an important role in hematopoietic cell signaling. The mid-sized isoform contains a short intracellular part without ITIMs, while the shortest isoform lacks the transmembrane segment and represents a potential soluble protein (2, 3).
