Description: The Akt signaling pathway contributes to the regulation of apoptosis after a variety of cell death signals. AKT1S1, also known as PRAS40, is a proline-rich substrate of the kinase AKT1 and is thought to play a role in neuroprotection mediated by nerve growth factor (NGF) after transient focal cerebral ischemia (1). AKT1S1 is also a substrate and potential regulator of mammalian target of rapamycin (mTOR), a serine/threonine kinase that regulates cell growth and cell cycle, and a negative regulator of autophagy (2). Treatment with the insulin-like growth factor-1 (IGF1) can indusce the phosphorylation of AKT1S1 via the PI3K/AKT signaling pathway in PC12 cells (3).

Catalog Number: (10800-552)
Supplier: Rockland Immunochemical
Description: Three distinct signaling pathways lead to programmed cell death (apoptosis). The death receptor and mitochondrion pathways are the mains, in which the key apoptotic proteases capase-8 and caspase-9, respectively, are involved. The endoplasmic reticulum (ER) stress is the third apoptotic pathway and caspase-12 is involved. Caspase-12 is localized to the ER but not to cytoplasm or mitochondrion. Caspase-12 is activated by ER stress, including disruption of ER calcium homeostasis, and mediates ER stress-induced apoptosis. Caspase-12 is co-localized to the ER with several proteins that are involved in Alzheimer's disease including gamma-secretase presenilin and beta-amyloid precursor protein (APP). Caspase-12 mediates cytotoxicity induced by amyloid-beta. Caspase-12 is ubiquitously expressed in mouse tissues.

Supplier: Rockland Immunochemical
Description: PTCHD2, also known as DISP3, is the third of three known homologs of the D. melanogaster protein Dispatched. It is a multi-transmembrane protein containing two PTCH/DISP domains and is thought to be involved in the release of lipid-anchored secreted proteins. Like DISP1 and DISP2, DISP3 has been implicated in signaling pathways during embryogenesis, tissue regeneration, and carcinogenesis. It is highly expressed in Purkinje cells, hippocampal neurons, and retinal ganglion cells. Recently, it has been shown that PTCHD2 localizes within the endoplasmic reticulum and colocalizes with cholesterol, and given that its expression is regulated by thyroid hormone (T3), it has been suggested that DISP3 may be a link between thyroid hormone and cholesterol metabolism.
