Description: CD2, also known as leukocyte function-associated antigen-2 (LFA-2), is a member of the immunoglobulin superfamily of cell adhesion molecules. It is expressed on the surface of >95% of thymocytes, and essentially all B cells, T cells and NK cells. In the mouse, CD2 is the receptor for CD48. CD2 is thought to play a major role in mediating T-cell adhesion, T-cell recognition/activation, T-cell signaling, and intrathymic differentiation.

Description: The IL-2 receptor is a complex of three distinct polypeptide chains: (i) the α chain which binds IL-2 with low affinity; (ii) the β chain that binds IL-2 with high affinity; and (iii) the common γ chain (γc) that does not bind IL-2. The high affinity receptor complex is an α/β/γ heterotrimer with a Kd of 1.3 x 10-11 M. In mouse spleen, CD122 is expresed on ~30% of CD8+ cells and all NK cells but <1% of B cells and CD4+ T lymphocytes. In the thymus, its expression is confined to CD4-CD8+ single positive and CD4-CD8- double negative cells. Cytoplasmic regions of the IL-2R β chain are involved in IL-2-mediated cellular signaling and, via the interaction of IL-2 and its receptor complex, may be involved in the generation and differentiation of T lymphocytes. The monoclonal antibody 5H4 does not block IL-2 binding.

Description: The BC96 monoclonal antibody reacts with human CD25, the 55 kDa interleukin-2 receptor α chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. By itself, CD25 binds IL-2 only with low affinity. However, CD25 associates with CD122 (IL-2 receptor β chain) and CD132 (common γ chain) to form the high affinity IL-2 receptor. CD25 plays a role in lymphocyte differentiating and activation/proliferation. CD25 is expressed on activated B cells, monocytes/macrophages, and T cells.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow.

Description: CD3ε, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chain ranging in size from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). It is expressed on all T cells of all mouse strains. CD3 plays a major role in signaling during antigen recognition, leading to T-cell activation. The monoclonal antibody C363.29B recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble C363.29B can activate primed and naïve T cell in vitro. Immobilized C363.2B9 monoclonal antibody can also activate both normal T lymphocytes and cloned T cell lines provided the appropriate accessory signals are present. The monoclonal antibody is cytolytic, easily used for cell surface staining, and a good immunoprecipitating antibody.

Description: CD80 (B7-1) is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed by dendritic cells, monocytes, and peritoneal macrophages. Expression of CD80 on B cells is upregulated after activation by various means including stimulation by LPS, IL-4, and surface Ig receptor crosslinking with peak expression occurring between 48 and 72 hours post-activation. The interaction of CD80 with its ligands, CD28 and CD152 (CTLA-4), provides a potent costimulatory signal for T cells activated through the CD3/T cell receptor complex. The monoclonal antibody 1G10 blocks binding of CTLA-4 to CD80 but it does not block stimulation of T cells by natural antigen-presenting cells.
