Description: The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 28-14-8 binds to the α3 domain of H-2Db in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Ld, but not Kd or Dd, and with H-2Dq and/or Lq. The antibody has been shown to block H-2Ld-specific and H-2Ld-restricted antigen-specific lysis of target cells by cytotoxic T lymphocytes but it does not block recognition of H-2Ld-positive target cells by Ly-6G2-positive NK cells.

Description: CD95, also known as Fas and APO-1, is a 45 kDa type I transmembrane glycoprotein and a member of the tumor necrosis factor receptor superfamily. It is expressed by activated lymphocytes, monocytes, neutrophils, fibroblasts, and cell lines. Fas ligand binding to CD95 induces apoptosis in activated mature lymphocytes thereby playing a role in maintaining peripheral tolerance. Crosslinking of CD95 by the monoclonal antibodies DX2 and DX3 delivers an apoptotic signal to Fas-sensitive cells indicating that these monoclonal antibodies recognize a functional epitope of CD95.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody 104 recognizes CD45 (leukocyte common antigen) on all leukocytes of mouse strains expressing the CD45.2 allotype (e.g., A, AKR, BALB/c, CBA/Ca, CBA/J, C3H/He, C57BL, C57BR, C57L, C58, DBA/1, DBA/2, NZB, SWR, 129).

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

Description: CD81, also known as TAPA-1, is an integral membrane protein expressed on a variety of cell types and has a high degree of sequence homology between human and mouse. CD81 is expressed on thymic stromal cells where it plays an important role in the transition of γδ+ T cells to more mature T cells with αβ T cell receptors. Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor which is composed of a TCR β chain and glycoprotein pTα. The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.

Description: CD56, an isoform of the neural cell adhesion molecule (NCAM), is a heavily glycosylated type I transmembrane protein. On human hematopoietic cells its expression is restricted to NK cells and a subpopulation of T lymphocytes (known as large granular lymphocytes) which demonstrate natural killer activity. Description: Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1-8). The two keratin types share only 30% sequence homology. Keratins demonstrate tissue- and differentiation-specific expression profiles. Cytokeratin 8 belongs to the type B (basic) subfamily of keratins and exists in combination with keratin 18. It is expressed in all simple type epithelia tissues (e.g., liver, pancreas, kidney, gut epithelial lining) but not in stratified squamous epithelia. Description: In the chicken, the CD8 molecule is present in two forms - (i) a homodimer of two α chains and (ii) a heterodimer of an α chain and a β chain. While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8α- and CD8β-chains, a relatively large proportion of the CD8+ TCRγδ cells in the spleens of embryos and young chicks express only the α-chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved but there is a population of TCRγδ CD8αβ cells in the chicken that is not found in rodents. Chicken CD8 is expressed on approximately 80% of thymocytes, 15% of blood mononuclear cells, and 50% of spleen cells but less than 1% of cells in the bursa and bone marrow. The monoclonal antibody CT-8 recognizes the CD8α chain and has been shown to react to a polymorphic determinant in turkey.
