Description: The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes, which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 34-1-2S binds to a common determinant in the α3 domains of H-2Kd and H-2Dd in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Kb.

Description: The αβ TCR is expressed on T lymphocytes of all mouse strains tested. The H57-597 monoclonal antibody does not react with γδ TCR-bearing cells. Plate-bound or soluble H57-597 activates αβ TCR-bearing T cells. In vitro and in vivo, H57-597 can induce immature thymocytes to undergo apoptosis. In vivo administration of H57-597 has been shown to deplete αβ TCR-bearing cells to near completion and prevent graft rejection.

Description: In the mouse, CD8 exists in two forms – (i) a CD8 heterodimer composed of an α chain (CD8α) and a β chain (CD8β); and (ii) a homodimer of two α chains. The heterodimer is found on the surface of essentially all thymocytes and the “suppressor/cytotoxic” subpopulation of mature T lymphocytes. Subsets of intestinal intraepithelial lymphocytes express CD8α without CD8β. It has been suggested that CD8+β- T cells mature extrathymically, while development of the CD8α+β+ population of T cells is thymus-dependent. CD8 acts as a coreceptor with MHC Class I-restricted T cell receptors in antigen recognition and positive selection of MHC class I-restricted CD8+ T cells. In vivo and in vitro treatment with the 53-6.7 monoclonal antibody effectively depletes CD8α+ cells. The 53-6.7 monoclonal antibody also blocks allogeneic help specific for class I MHC antigens and T cell responses to IL-2.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.

Description: The monoclonal antibody TCR-2 precipitates a CD3-associated heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 34 kDa and 29 kDa. In the chicken, two distinct subpopulations of αβ T cells appear in the thymus subsequent to the appearance of γδ T cells. These subpopulations, originally denoted as TCR2 and TCR3, arise sequentially in the thymus during ontogeny and are now known to represent two distinct Vβ families, Vβ1 and Vβ2, respectively. The TCR-2 monoclonal antibody reacts with approximately 40% of thymocytes, 40-50% of blood mononuclear cells, and 40% of splenocytes in the chicken. Two-color immunofluorescence has revealed that most of the TCR2+ thymocytes express both CD4 and CD8 antigens. The TCR2+ cells in blood were found to express either CD4 (74 ± 2%) or CD8 (26 ± 4%). TCR2+ cells in the spleen also express either CD4 (37 ± 1%) or CD8 (64 ± 4%). Surprisingly, a relatively large subpopulation of CD8+ cells in the spleen are negative for TCR2. This observation led to the demonstration that 71 ± 6% of the TCRγδ+ cells in the spleen express CD8.
