Description: CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins. Murine CD30 is expressed predominantly in the thymus and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A. In anti-CD3ε-activated spleen cells, CD30 is expressed primarily on the surface of CD8+ T cells with peak expression on days 4 and 5. Stimulation of CD30+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN-γ production. While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.

Description: The monoclonal antibody 14-4-4S reacts with the I-Ek class II alloantigen on cells from mice of the H-2d, H-2p, and H-2r haplotypes. Cells from mice of the H-2b, H-2f, H-2q, and H-2s haplotypes do not express I-E antigen. The antibody has been reported to cross-react with the rat MHC class II alloantigen RT1D.

Description: CD103 is a member of the integrin series of adhesion molecules. This antigen defines a developmentally important subset of T cells, namely mucosal T cells including all IEL (intraepithelial lymphocytes) and ~20% of lamina propria T cells. Expression of CD103 is more restricted outside these mucosal organs appearing at lower levels on T cell subsets of the lymph node, dendritic epidermis, and periphery. In non-epithelial CD103+ T cells there is a bias toward expression on CD8+ cells. The 2E7 monoclonal antibody exhibits antigen immunoprecipitation patterns similar if not identical to the rat anti-mouse CD103 monoclonal antibody M290. 2E7 is reported to have signal-inducing activity in a redirected lysis assay and to costimulate IEL and CD8+ lymph node cells in conjunction with anti-TCR in vitro which runs counter to the usual inhibition of CTL by anti-integrins. This costimulatory activity is also shared by the monoclonal antibody M290. The property of costimulation is not unique to the mouse since the combination of anti-human αIEL plus anti-TCR will produce similar proliferation in human cells with the exception that CD8+ cells are not stimulated.

Description: CD44, also known as phagocytic glycoprotein-1 (Pgp-1), exists as a large number of different isoforms resulting from alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type I transmembrane protein. Other variable isoforms containing glycosaminoglycans have molecular weights ranging from 110 to 250 kDa and are widely expressed on hematopoietic and non-hematopoietic cells. CD44 contributes to the adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix. It also plays a functional role in cell migration, lymphocyte homing, and adhesion during hematopoiesis and lymphocyte activation.

Description: The monoclonal antibody TCR-1 precipitates a heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 35 kDa and 32 kDa. Unlike human and mouse in which γδ cells comprise a minor subset of T lymphocytes in the circulation, the chicken has a relatively large subset of γδ T cells. The frequency of TCRγδ cells is typically 20-25% of the total blood T cells but may reach approximately 50% in chickens of six- months of age. The majority of TCRγδ+ cells in the thymus and blood are CD4-CD8- although a small subset of them may express CD8 or CD4 coreceptors. However, when the TCRγδ+ cells migrate into the spleen and intestine most of them begin to express CD8. Although the biological function of γδ T cells is unclear, they are clearly capable of cytotoxic activity in vitro. The CD8+ γδ T cells may also be involved in down-regulation of the immune response. They cannot, however, induce a graft-vs-host (GVH) reactions.

Description: CD3, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chains ranging from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). CD3 is expressed on 70-80% of normal peripheral blood lymphocytes and on 10-20% of thymocytes. It plays a major role in signaling during antigen recognition, leading to T cell activation. The monoclonal antibody UCHT1 reacts with the 20 kDa ε chain of the CD3/TCR complex.

Description: Protein tyrosine residues are phosphorylated as a result of intracellular protein kinase activation (e.g., via growth factors) during normal growth and development and in oncogenesis. The most abundant population of target proteins for tyrosine phosphorylation is cell surface glycoproteins. Antibodies to phosphotyrosine enable the detection, isolation, and characterization of proteins containing phosphotyrosine. The monoclonal antibody PY20 prevents internalization of activated receptors (e.g., EGFR) when microinjected into cells. The affinity of PY20 for phosphotyrosine is approximately 10-6 to 10-7 M. PY20 binding to phosphorylated tyrosines can be inhibited by free phosphotyrosine and phenylphosphate but not by phosphoserine, phosphothreonine, or free phosphate.

Description: ZAP-70 is a protein tyrosine kinase (PTK) with the primary role of catalyzing the phosphorylation of tyrosine residues in cellular proteins, and thus, contributes significantly to the initiation of cell signaling. ZAP-70 is normally expressed on T cell and NK cells.

Description: Feline CD5, a member of the scavenger receptor family of cell surface receptors, is a type I transmembrane glycoprotein that is expressed on most thymocytes and mature T lymphocytes. CD5 is thought to function in both cell-cell recognition and signal transduction. The monoclonal antibody f43 is useful as a pan T cell marker.

Description: The monoclonal antibody 5H10.27 reacts with the α5 chain (VLA-5) of the CD49e/CD29 heterodimeric fibronectin receptor. It is expressed on thymocytes, activated T cells, mast cells, and a variety of mouse cell lines. Soluble 5H10.27 antibody inhibits VLA-5-mediated functions in vitro. Immobilized 5H10.27 has also been demonstrated to costimulate the proliferative response of CD8+ T cells to plate-bound anti-CD3 monoclonal antibody.
