Description: CD44, also known as phagocytic glycoprotein-1 (Pgp-1), exists as a large number of different isoforms resulting from alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type I transmembrane protein. Other variable isoforms containing glycosaminoglycans have molecular weights ranging from 110 to 250 kDa and are widely expressed on hematopoietic and non-hematopoietic cells. CD44 contributes to the adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix. It also plays a functional role in cell migration, lymphocyte homing, and adhesion during hematopoiesis and lymphocyte activation.

Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

The monoclonal antibody DX9 is expressed on a subset of CD3-, CD56+ NK cells isolated from peripheral blood at varying levels (<0.1% to 61%) among individuals but not on thymocytes, B lymphocytes, granulocytes, or monocytes. Interactions with CD158e1 and certain MHC HLA-B antigens on a target cell inhibits cytotoxicity, possibly preventing target cell lysis and death. Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.
