Description: CD43, a member of the sialomucin family of cell surface receptors, is a 95-135 kDa type I transmembrane glycoprotein whose variability in molecular weight is dependent upon the nature of the extracellular O-glycans. It is the major sialoglycoprotein on thymocytes, T cells, and neutrophils. It is also present on activated B cells, plasma cells, NK cells, granulocytes, monocytes, macrophages, platelets, and bone marrow hematopoietic stem cells. CD43, which binds to ICAM-1 (CD54), appears to function as an anti-adhesion molecule, inhibiting T cell interactions including T cell killing, and by increasing the threshold for T cell activation.

Description: In the chicken, the CD8 molecule is present in two forms - (i) a homodimer of two α chains and (ii) a heterodimer of an α chain and a β chain. While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8α- and CD8β-chains, a relatively large proportion of the CD8+ TCR-γδ cells in the spleens of embryos and young chicks express only the α-chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved but there is a population of TCR-γδ CD8αβ cells in the chicken that is not found in rodents. Chicken CD8 is expressed on approximately 80% of thymocytes, 45% of blood mononuclear cells, and 50% of spleen cells but less than 1% of cells in the bursa and bone marrow. The monoclonal antibody EP42 recognizes the CD8β chain.

Description: CD154, formerly known as CD40 ligand and gp39, is a type II integral membrane protein and a member of the tumor necrosis factor (TNF) family of ligands. It is an important accessory molecule in T cell-B cell costimulatory interactions and is expressed predominantly on activated CD4+ T lymphocytes. It is also present on the surface of activated Th0, Th1, and Th2 T cell clones. Its expression is transient and cyclosporin-sensitive. The MR1 monoclonal antibody binds to murine CD154 with high affinity, blocks binding to CD40, and blocks CD154 function. Administration of this antibody to mice blocks the ability to mount primary and secondary immune responses to TD antigens yet does not alter the immune response to TI antigens.

Description: The CD117 (c-Kit) antigen is the cell-surface receptor for stem cell factor (SCF). Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: CD54, also known as intercellular cell adhesion molecule-1 (ICAM-1), is a 90 kDa type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It exhibits a wide tissue distribution being expressed on both hematopoietic and non-hematopoietic cells. ICAM-1 expression on leukocytes is low but is rapidly upregulated upon activation. Expression on endothelium and other non-hematopoietic cells is strongly upregulated by inflammatory mediators. The ligands of CD54 are CD11a/LFA-1α and CD11b/Mac-1α. Endothelial CD54 contributes to the extravasation of leukocytes from blood vessels particularly in areas of inflammation. CD54 on antigen-presenting cells (APC) contributes to antigen-specific T cell activation presumably by enhancing interactions between T cells and APC.

Description: CD62E represents the 97 kDa type I transmembrane glycoprotein E-selectin, also known as endothelial-leukocyte adhesion molecule-1 (ELAM-1). It is expressed on cytokine-stimulated endothelium, weakly on unactivated endothelium, and is absent on leukocytes. Like other selectins, CD62E is an adhesion molecule that contributes to the initial attachment and rolling of leukocytes on endothelial surfaces, a prerequisite for leukocyte extravasation into tissues. Description: CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion. CD107a is expressed by activated T cells, macrophages, dendritic cells, activated platelets, tonsillar epithelium, and some tumor cell lines, including U937 and KG1a. It is also a widely expressed intracellular antigen. LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.
