Description: In the mouse, CD8 exists in two forms – (i) a CD8 heterodimer composed of an α chain (CD8α) and a β chain (CD8β); and (ii) a homodimer of two α chains. The heterodimer is found on the surface of essentially all thymocytes and the “suppressor/cytotoxic” subpopulation of mature T lymphocytes. Subsets of intestinal intraepithelial lymphocytes express CD8α without CD8β. It has been suggested that CD8+β- T cells mature extrathymically, while development of the CD8α+β+ population of T cells is thymus-dependent. CD8 acts as a coreceptor with MHC Class I-restricted T cell receptors in antigen recognition and positive selection of MHC class I-restricted CD8+ T cells. In vivo and in vitro treatment with the 53-6.7 monoclonal antibody effectively depletes CD8α+ cells. The 53-6.7 monoclonal antibody also blocks allogeneic help specific for class I MHC antigens and T cell responses to IL-2.

CD49b associates noncovalently with CD29, the integrin β1 subunit, to form the heterodimer integrin α2β1 that binds laminin and collagen. It is expressed on B and T lymphocytes, NK cells, monocytes, platelets, fibroblasts, and long-term cultured T cells. CD49b/CD29 mediates the Mg++-dependent adhesion of platelets to collagen. It may also be utilized by fibroblasts for the reorganization of the collagen matrix during wound healing.

Description: The immunoglobulin heavy chain binding protein BiP is a member of the hsp70 family of heat shock proteins and is identical to the glucose regulated protein GRP78. While BiP was originally described for its function in B cells, it is now known to be distributed in a variety of tissues, if not ubiquitous. The highly conserved hsp70 proteins have an essential physiological role in stress responses and as “molecular chaperones” which are responsible for a variety of functions such as protein transport, prevention of protein toxicity and direction of protein folding. With regard to its immunological role, BiP is a component of the endoplasmic reticulum and binds free intracellular heavy chains in nonsecreting pre-B cell lines (μ+,L-) or incompletely assembled Ig precursors in H+L+ secreting hybridomas and myelomas. In the absence of light chain synthesis, heavy chains remain associated with BiP and are not secreted. BiP is an ATP binding protein and the dissociation of the BiP-heavy chain complex is probably driven by the ATPase activity attributed to BiP. The monoclonal antibody 76-E6 recognizes a conserved epitope localized within the region of amino acids 497 to 581 of BiP.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

Description: The monoclonal antibody 20d5 reacts with an epitope common to the NKG2A, NKG2C and NKG2E isoforms of the NKG2 heterodimer, a member of the c-type lectin family of inhibitory receptors. Mouse NKG2 is a natural killer (NK) cell receptor for the non-classical MHC class I molecule Qa-1b. NKG2 is expressed on NK cells, lymphokine-activated killer (LAK) T cells, and some CD8+ memory T cells. Two-color immunofluorescent staining with the 20d5 antibody reveals two distinct NKG2+ phenotypes within the NK1.1+CD3- spleen cell population: NKG2hi (50%) and NKG2low (50%). With the exception of DBA/2J mice, NKG2 is expressed on NK1.1+ cells from all mouse strains tested (C57BL/6, BALB/c, 129/J, C3H.SW, AKR/J, SJL).

Description: CD90 (Thy-1) is a GPI-anchored molecule and one of the smallest members of the immunoglobulin superfamily consisting of a single V-set domain. The Thy-1.2 alloantigen is expressed on all thymocytes, peripheral T lymphocytes, and some intraepithelial T cells of most mouse strains. It is also expressed in the brain and at varying levels on other non-lymphoid tissues. It is detected in the cytoplasm early in B cell development (late pro-B cell stage), appears on the cell surface concomitant with IgD, and is found on most IgM+IgD+ mature B lymphocytes. Expression is lost with terminal differentiation of B cells and is absent on plasma cells. Activation of B cells via cross-linking of surface Ig increases CD22 expression. CD22 associates with the B cell receptor (BCR) complex and mediates intercellular adhesion. Its intracellular domain is phosphorylated after antigen receptor cross-linking and is involved in negative regulation of B cell activation.
