Description: The mammalian Target of Rapamycin (TOR, also known as mTOR) is an evolutionarily conserved serine/threonine kinase that regulates cell growth and cell cycle through its ability to integrate signals from nutrient levels and growth factors. It was initially discovered as a kinase whose ability to stimulate T cell proliferation in response to IL-2 could be inhibited by the immunosuppressive drug rapamycin. Rapamycin inhibits TOR in other cell types resulting in reduced cell growth and reduced rates of cell cycle and cell proliferation. TOR is normally associated with the regulatory proteins RAPTOR and GbetaL. Its downstream targets are thought to be the ribosomal protein S6 kinases and the eukaryotic initiation factor 4E binding proteins (4EBPs). Regulation of these protein families allows TOR to control protein biosynthesis.

RIM1 and RIM2 proteins are expressed in overlapping but distinct patterns throughout the brain. While the ablation of either gene was not lethal in mice, the deletion of both resulted in postnatal mortality. This lethality is due to a defect in neurotransmitter release; synapses without RIM proteins can still release neurotransmitters but are unable to do so in response to normal Ca2+ triggers. Deprivation of BCAS2 through RNA inhibition induces apoptosis in p53-wild type cells, but causes G2-M arrest in p53-null or -mutant cells; this effect was reversed with the expression of ectopic BCAS2. Description: Alpha-tubulin belongs to the tubulin superfamily, which is composed of six distinct families. Along with beta-tubulins, alpha-tubulins are the major components of microtubules. These microtubules are involved in a wide variety of cellular activities ranging from mitosis and transport events to cell movement and the maintenance of cell shape. Alpha- and beta-tubulin dimers are assembled to 13 protofilaments that form a microtubule of 22-nm diameter. Tyrosine ligase adds a C-terminal tyrosine to monomeric alpha-tubulin. Assembled microtubules can again be detyrosinated by a cytoskeleton-associated carboxypeptidase. Another post-translational modification of detyrosinated alpha-tubulin is C-terminal polyglutamylation, which is characteristic of microtubules in neuronal cells and the mitotic spindle.
