Description: The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.

Description: CD69, also known as very early activation (VEA) antigen, is a disulfide-linked transmembrane homodimer whose differentially glycosylated subunits range from 35-39 kDa. It is a C-type lectin most closely related to the NKR-P1 and Ly-49 NK cell-activation molecules. CD69 is widely expressed on hematopoietic cells, including lymphocytes, neutrophils and eosinophils. Although not detectable on resting lymphocytes, its expression is rapidly upregulated upon activation of T, B and NK cells, and neutrophils. Constitutive expression of CD69 on subsets of thymocytes suggests that it may be involved in regulation of developmental events in addition to its role in activation of a variety of hematopoietic cells. The monoclonal antibody H1.2F3 augments PMA-induced T-cell proliferation and induces redirected lysis of Fc receptor-bearing target cells by NK cells.

Description: The monoclonal antibody TCR-1 precipitates a heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 35 kDa and 32 kDa. Unlike human and mouse in which γδ cells comprise a minor subset of T lymphocytes in the circulation, the chicken has a relatively large subset of γδ T cells. The frequency of TCRγδ cells is typically 20-25% of the total blood T cells but may reach approximately 50% in chickens of six- months of age. The majority of TCRγδ+ cells in the thymus and blood are CD4-CD8- although a small subset of them may express CD8 or CD4 coreceptors. However, when the TCRγδ+ cells migrate into the spleen and intestine most of them begin to express CD8. Although the biological function of γδ T cells is unclear, they are clearly capable of cytotoxic activity in vitro. The CD8+ γδ T cells may also be involved in down-regulation of the immune response. They cannot, however, induce a graft-vs-host (GVH) reactions.

Description: CD106, also known as VCAM-1, is an adhesion molecule and a major mediator of the inflammatory response. It is expressed on activated microvascular endothelial cells in response to signals arising from immune responses in infection, graft rejection, tumor recognition and killing. The complementary binding ligand for VCAM-1 is VLA-4/CD49d. In addition to VCAM-1, VLA-4 also recognizes the extracellular matrix molecule fibronectin. This pairing of VCAM-1 and VLA-4 is able to provide a second signal (e.g., non-antigen specific) for T cell stimulation, such as that seen in transplantation. The monoclonal antibody MK-2 has been used in transplant studies to suppress cardiac rejection and induce long-term cardiac graft survival. In addition to inflammatory responses, VCAM-1 has a significant role in hemopoiesis through its ability to retain lymphocyte and myeloid precursors on stromal cells in the marrow and lymphoid organs. CD106/VCAM-1 exists as an integral membrane protein. The M/K-2 monoclonal antibody immunoprecipitates a peptide that gives a single band on SDS-PAGE gels with an apparent Mr of ~100 kDa under reducing conditions and 92 kDa under non-reducing conditions.
