Description: NK1.1, a member of the NKR-P1 family of cell surface receptors, is a type II integral membrane glycoprotein with a C-type lectin domain. It is expressed as a disulfide-linked homodimer on all NK cells as well as subsets of thymocytes and peripheral T lymphocytes in selected strains of mice (e.g., C57BL/6, NZB, and CE). NK1.1 mediates cellular activation and differentiation, and is thought to have a particular role in generating Th2 cells. This product does not react with NK cells of BALB/c mice.

Description: CD30 is a type I transmembrane protein and a member of the tumor necrosis factor receptor superfamily of proteins. Murine CD30 is expressed predominantly in the thymus and it is inducible in mouse splenocytes stimulated with pokeweed mitogen or concanavalin A. In anti-CD3ε-activated spleen cells, CD30 is expressed primarily on the surface of CD8+ T cells with peak expression on days 4 and 5. Stimulation of CD30+ CTL lines with plate-bound anti-CD30 directly signals IL-5 but not IFN-γ production. While these studies demonstrate that CD30 directs cytokine secretion and suggest that CD30 may play a pivotal role in the pattern of cytokine production by T cells, the precise roles of CD30 and its ligand (CD153) in T-cell development have not been clearly defined.

Description: The CD117 (c-Kit) antigen is the cell-surface receptor for stem cell factor (SCF). Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: ChT1, a member of the Ig superfamily with one V-like and one C2-like domain, is a T cell antigen that is expressed on the surface of embryonic thymocytes (day 10). In young chickens about 90% of the thymocytes as well as a subpopulation of peripheral lymphocytes, which represent recent thymic emigrants are ChT1 positive. Expression in the periphery declines with age and, in correlation with partial thymectomy, indicates that ChT1 can be used as an accurate marker for studying thymic function. The monoclonal antibody CT1 also recognizes the ChT1 antigen in quail cortical thymocytes.

Description: Chicken Bu-1a, a product of the Bu-1a allele, is expressed on the cell surface as a disulfide-linked homodimer. In 10-week-old H.B15 and H.B2 chickens, Bu-1 is found on 85-90% of bursal cells, 2-8% of thymocytes, 15-27% of spleen cells, and 2-18% of peripheral blood cells. It is also expressed on subsets of macrophages and monocytes but not on granulocytes, erythrocytes, or thrombocytes. In chickens heterozygous for the Bu-1 alleles (Bu-1a/b), Bu-1a does not exhibit allelic exclusion. The monoclonal antibody 21-1A4 does not react with cells from CHA and H.B14ab strains by immunofluorescence.

Description: CD106, also known as VCAM-1, is an adhesion molecule and a major mediator of the inflammatory response. It is expressed on activated microvascular endothelial cells in response to signals arising from immune responses in infection, graft rejection, tumor recognition and killing. The complementary binding ligand for VCAM-1 is VLA-4/CD49d. In addition to VCAM-1, VLA-4 also recognizes the extracellular matrix molecule fibronectin. This pairing of VCAM-1 and VLA-4 is able to provide a second signal (e.g., non-antigen specific) for T cell stimulation, such as that seen in transplantation. The monoclonal antibody MK-2 has been used in transplant studies to suppress cardiac rejection and induce long-term cardiac graft survival. In addition to inflammatory responses, VCAM-1 has a significant role in hemopoiesis through its ability to retain lymphocyte and myeloid precursors on stromal cells in the marrow and lymphoid organs. CD106/VCAM-1 exists as an integral membrane protein. The M/K-2 monoclonal antibody immunoprecipitates a peptide that gives a single band on SDS-PAGE gels with an apparent Mr of ~100 kDa under reducing conditions and 92 kDa under non-reducing conditions.

Description: Glial fibrillary acid protein (GFAP) is a member of the type III intermediate filament family of proteins. GFAP is heavily expressed in astrocytes and certain other astroglia in the central nervous system, in satellite cells in peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. It is closely related to its non-epithelial family members, vimentin, desmin, and peripherin, which are all involved in the structure and functions of the cell’s cytoskeleton. GFAP is thought to help to maintain astrocyte mechanical strength, as well as the shape of cell; however, its precise function remains poorly understood. In adults, GFAP levels increase in response to the proliferation of astrocytes associated with Alzheimer’s disease, epilepsy, and multiple sclerosis. Antibodies specific for GFAP are useful as markers of astrocytic cells and neural stem cells as well as for distinguishing neoplasms of astrocytic origin from other neoplasms in the central nervous system.
