Description: In the mouse, CD8 exists in two forms – (i) a CD8 heterodimer composed of an α chain (CD8α) and a β chain (CD8β); and (ii) a homodimer of two α chains. The heterodimer is found on the surface of essentially all thymocytes and the “suppressor/cytotoxic” subpopulation of mature T lymphocytes. Subsets of intestinal intraepithelial lymphocytes express CD8α without CD8β. It has been suggested that CD8+β- T cells mature extrathymically, while development of the CD8α+β+ population of T cells is thymus-dependent. CD8 acts as a coreceptor with MHC Class I-restricted T cell receptors in antigen recognition and positive selection of MHC class I-restricted CD8+ T cells. In vivo and in vitro treatment with the 53-6.7 monoclonal antibody effectively depletes CD8α+ cells. The 53-6.7 monoclonal antibody also blocks allogeneic help specific for class I MHC antigens and T cell responses to IL-2.

Description: CD14 is a 55 kDa glycosphosphatidylinositol (GPI)-anchored single chain glycoprotein. It is expressed on monocytes, macrophages, dendritic cells, and Langerhans cells. It is also weakly expressed on neutrophils. CD14 appears to be involved in clearance of gram-negative bacteria via its high affinity binding to LPS-LPB complexes.

Description: The monoclonal antibody TCR-1 precipitates a heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 35 kDa and 32 kDa. Unlike human and mouse in which γδ cells comprise a minor subset of T lymphocytes in the circulation, the chicken has a relatively large subset of γδ T cells. The frequency of TCRγδ cells is typically 20-25% of the total blood T cells but may reach approximately 50% in chickens of six- months of age. The majority of TCRγδ+ cells in the thymus and blood are CD4-CD8- although a small subset of them may express CD8 or CD4 coreceptors. However, when the TCRγδ+ cells migrate into the spleen and intestine most of them begin to express CD8. Although the biological function of γδ T cells is unclear, they are clearly capable of cytotoxic activity in vitro. The CD8+ γδ T cells may also be involved in down-regulation of the immune response. They cannot, however, induce a graft-vs-host (GVH) reactions.

Description: The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.

Description: CD45RC represents an exon C-dependent isoform of CD45 glycoprotein. It is expressed at high density on all B lymphocytes, at intermediate density on most CD8+ splenic T cells, and on two subpopulations of CD4+ splenic T cells with high and low density of expression. CD45RC is also present on a small subset of thymocytes.

Description: CD3ε, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chains ranging in size from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). It is expressed on all T cells of all mouse strains. CD3 plays a major role in signaling during antigen recognition, leading to T-cell activation. The 145-2C11 monoclonal antibody recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble 145-2C11 can activate primed and naïve T cell in vitro. 145-2C11 can also induce redirected lysis of specific target cells by CTL clones and it can block lysis of specific target cells by antigen-specific CTL’s. Immobilized 145-2C11 can activate both normal T lymphocytes and cloned T cell lines. Under certain conditions, T cell activation by 145-2C11 may result in apoptotic cell death.

Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. Expression of CD25 on activated T lymphocytes is transitory and endogenously regulated. CD25 is expressed on precursor B cells in bone marrow. Its expression is initiated by functional rearrangement and expression of IgM heavy chain genes and is down-regulated when immature B cells mature and express IgD. It is expressed at a higher level on CD4+CD8+ thymocytes. It is also expressed on cultured epidermal Langerhans cells. The biochemical features of murine CD25 have been characterized in detail. The 3C7 monoclonal antibody reacts with an epitope of CD25 which is distinct from that recognized by clone 7D4. 3C7, used in combination with 7D4 in culture, results in higher levels of inhibition of proliferation driven by IL-2 and generation of alloreactive CTL than either of these monoclonal antibodies alone.

Description: CD19 is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily. It is expressed at all stages of B cell differentiation except terminally differentiated plasma cells. It is also present on the cell surface of follicular dendritic cells. CD19 associates with CD21, CD81, Leu 13 and/or MHC Class II molecules to form a signal transduction complex on the B cell surface. This signaling complex modulates the activation threshold for the B cell antigen receptor (BCR).
