Description: CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. Description: CD107b, also known as lysosomal-associated membrane protein 2 (LAMP-2), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains and is a ligand for E-selectin-mediated cell adhesion. CD107b is expressed in lysosomal/endosomal membranes and on the surface of activated platelets and some tumor cell lines including U937 and KG1a. LAMP-2 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

Description: The monoclonal antibody DX5 reacts with CD49b, also known as very late antigen-2 (VLA-2) and α2 integrin. The antibody stains the majority of NK cells and a small subpopulation of T cells in all mouse strains tested (e.g., A/J, AKR, BALB/c, C3H/HeJ, C57BL/6, C57BL/10, C57BR, C58, CBA/Ca, CBA/J, DBA/1, DBA/2, SJL, SWR). Three-color flow cytometry analysis has revealed that most cells that express NK-1.1 (NKR-P1C) also express CD49b. However, small subsets of DX5+NK-1.1- and DX5-NK-1.1+ cells can be found, especially among the CD3+ population of T lymphocytes. NK cells cultured in the presence of IL-2 progressively lose reactivity with DX5 as a consequence of cellular proliferation. Therefore, DX5 can be used to define functionally distinct subsets of murine NK cells. The monoclonal antibody DX5 has not been demonstrated to have activating or blocking activity.

Description: CD44 represents a family of type I transmembrane glycoproteins expressed on many cell types such as B cells, monocytes, macrophages, and variable subsets of thymocytes and peripheral T cells. CD44 primarily functions as an adhesion molecule. It has been implicated in various processes, such as hematopoiesis, lymphocyte homing, leukocyte activation, tumor metastasis and development. The KM201 monoclonal antibody reacts with an epitope very close to the hyaluronate binding domain on the CD44 molecule. KM201 can inhibit hyaluronate-dependent cell aggregation, prevent lympho-hemopoiesis in both Dexter and Whitlock-Witte cultures, prevent the earliest intrathymic precursors from homing to the thymus, and costimulate the activation of freshly purified splenic CD4+ T cells.

Description: CD44, also known as phagocytic glycoprotein-1 (Pgp-1), exists as a large number of different isoforms resulting from alternative RNA splicing. The major isoform expressed on lymphocytes, myeloid cells, and erythrocytes is a glycosylated type I transmembrane protein. Other variable isoforms containing glycosaminoglycans have molecular weights ranging from 110 to 250 kDa and are widely expressed on hematopoietic and non-hematopoietic cells. CD44 contributes to the adhesion of leukocytes to endothelial cells, stromal cells, and the extracellular matrix. It also plays a functional role in cell migration, lymphocyte homing, and adhesion during hematopoiesis and lymphocyte activation.

Description: Glial fibrillary acid protein (GFAP) is a member of the type III intermediate filament family of proteins. GFAP is heavily expressed in astrocytes and certain other astroglia in the central nervous system, in satellite cells in peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. It is closely related to its non-epithelial family members, vimentin, desmin, and peripherin, which are all involved in the structure and functions of the cell’s cytoskeleton. GFAP is thought to help to maintain astrocyte mechanical strength, as well as the shape of cell; however, its precise function remains poorly understood. In adults, GFAP levels increase in response to the proliferation of astrocytes associated with Alzheimer’s disease, epilepsy, and multiple sclerosis. Antibodies specific for GFAP are useful as markers of astrocytic cells and neural stem cells as well as for distinguishing neoplasms of astrocytic origin from other neoplasms in the central nervous system.
