Description: CD3ε, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chain ranging in size from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). It is expressed on all T cells of all mouse strains. CD3 plays a major role in signaling during antigen recognition, leading to T-cell activation. The monoclonal antibody C363.29B recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble C363.29B can activate primed and naïve T cell in vitro. Immobilized C363.2B9 monoclonal antibody can also activate both normal T lymphocytes and cloned T cell lines provided the appropriate accessory signals are present. The monoclonal antibody is cytolytic, easily used for cell surface staining, and a good immunoprecipitating antibody.

Description: BP-1, also known as Ly-51 and 6C3 antigen, is a type II homodimeric transmembrane glycoprotein formed by two disulfide-linked chains of 140 kDa. Originally identified on the surface of pre-B cell lymphomas the cDNA encoding BP-1 has been cloned and found to represent aminopeptidase A. It is expressed at high levels on bone marrow stromal cell lines which support in vitro B lymphopoiesis and on a wide variety of mouse tissues known to possess aminopeptidase activity. Subsets of normal bone marrow pre-B and B cells express low levels of BP-1 which is rapidly upregulated on pre-B cells in the presence of IL-7. The BP-1 marker is commonly used to identify B cells in different stages of development.

Description: CD24 is expressed in B cells until their terminal differentiation into plasma cells. It has been used widely as a marker to fractionate different B-lineage differentiation stages. CD24 has been found on a variety of other hematopoietic and neural cell types and is now classified as a costimulatory molecule. The CD24 molecule, also known as HSA, is composed mostly of carbohydrate with a protein core of only 31-35 amino acids in the human and 27 amino acids in the mouse. CD24 is now included in the large group of phosphatidylinositol-anchored membrane proteins. Studies with purified CD24 applied to latex beads have demonstrated a self-binding property suggesting that the molecule is capable of promoting homotypic aggregation, binding, and cell adhesion. This is supported by earlier observations of the blockade of B cell lymphoblast aggregation with an anti-CD24 monoclonal antibody. Recent studies of murine CD24 have assigned a role for CD24 as a ligand for P-selectin, a calcium-dependent lectin.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation.
