Description: CD3ε, a member of the immunoglobulin superfamily of cell surface receptors, is comprised of five invariable chain ranging in size from 16-28 kDa and is closely associated with the T cell antigen receptor (TCR). It is expressed on all T cells of all mouse strains. CD3 plays a major role in signaling during antigen recognition, leading to T-cell activation. The monoclonal antibody C363.29B recognizes an epitope on the 25 kDa ε chain of the CD3/TCR complex. In the presence of Fc receptor-bearing accessory cells, soluble C363.29B can activate primed and naïve T cell in vitro. Immobilized C363.2B9 monoclonal antibody can also activate both normal T lymphocytes and cloned T cell lines provided the appropriate accessory signals are present. The monoclonal antibody is cytolytic, easily used for cell surface staining, and a good immunoprecipitating antibody.

Description: CD80 (B7-1) is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed by dendritic cells, monocytes, and peritoneal macrophages. Expression of CD80 on B cells is upregulated after activation by various means including stimulation by LPS, IL-4, and surface Ig receptor crosslinking with peak expression occurring between 48 and 72 hours post-activation. The interaction of CD80 with its ligands, CD28 and CD152 (CTLA-4), provides a potent costimulatory signal for T cells activated through the CD3/T cell receptor complex. The monoclonal antibody 1G10 blocks binding of CTLA-4 to CD80 but it does not block stimulation of T cells by natural antigen-presenting cells.

Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description: Fas ligand (CD178) is a 40 kD type II transmembrane protein that is a member of the tumor necrosis factor superfamily. This well characterized potent apoptotic factor is utilized by cytotoxic T cells and natural killer cells to selectively kill virus infected and tumorigenic cells. Moreover, studies indicate that Fas ligand is an important regulator in immune homeostasis where it has been shown to down-regulate immune responses during activation-induced cell death. Fas ligand initiates apoptosis by binding to its receptor, CD95, on target cells which facilitates recruitment of numerous signaling proteins to form a death inducing signaling complex. This signaling complex is then believed to propagate the apoptotic signal through the recruitment and activation of caspase-8. The SB93a monoclonal antibody detects a band at ~40 kDa corresponding to the transmembrane/insoluble form of Fas ligand.

Description: CD154, formerly known as CD40 ligand and gp39, is a type II integral membrane protein and a member of the tumor necrosis factor (TNF) family of ligands. It is an important accessory molecule in T cell-B cell costimulatory interactions and is expressed predominantly on activated CD4+ T lymphocytes. It is also present on the surface of activated Th0, Th1, and Th2 T cell clones. Its expression is transient and cyclosporin-sensitive. The MR1 monoclonal antibody binds to murine CD154 with high affinity, blocks binding to CD40, and blocks CD154 function. Administration of this antibody to mice blocks the ability to mount primary and secondary immune responses to TD antigens yet does not alter the immune response to TI antigens.
