Description: Chicken Bu-1, a product of the Bu-1a and Bu-1b alleles, is a type I transmembrane glycosylated disulfide-linked homodimer. The monoclonal antibody AV20 recognizes a monomorphic determinant on the Bu-1 B cell-associated alloantigens of both RPL 6(3) (Bu-1a) and 7(2) (Bu-1b) lines of inbred chickens. The antibody also identifies B cells in partially inbred birds. Bu-1 is found on 85-90% of bursal cells, 2-8% of thymocytes, 15-27% of spleen cells, and 2-18% of peripheral blood cells. It is also expressed on subsets of macrophages and monocytes but not on granulocytes, erythrocytes, or thrombocytes. The monoclonal antibody AV20 does not cross react with turkey cells when analyzed by flow cytometry.

Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description: CD81, also known as TAPA-1, is an integral membrane protein expressed on a variety of cell types and has a high degree of sequence homology between human and mouse. CD81 is expressed on thymic stromal cells where it plays an important role in the transition of γδ+ T cells to more mature T cells with αβ T cell receptors. Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor which is composed of a TCR β chain and glycoprotein pTα. The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro.

Description: The IL-2 receptor is a complex of three distinct polypeptide chains: (i) the α chain which binds IL-2 with low affinity; (ii) the β chain that binds IL-2 with high affinity; and (iii) the common γ chain (γc) that does not bind IL-2. CD49b associates noncovalently with CD29, the integrin β1 subunit, to form the heterodimer integrin α2β1 that binds laminin and collagen. It is expressed on B and T lymphocytes, NK cells, monocytes, platelets, fibroblasts, and long-term cultured T cells. CD49b/CD29 mediates the Mg++-dependent adhesion of platelets to collagen. It may also be utilized by fibroblasts for the reorganization of the collagen matrix during wound healing.

Description: In the chicken, the CD8 molecule is present in two forms - (i) a homodimer of two α chains and (ii) a heterodimer of an α chain and a β chain. Chicken CD8 is expressed on approximately 80% of thymocytes, 15% of blood mononuclear cells and 50% of spleen cells but less than 1% of cells in the bursa and bone marrow. While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8α- and CD8β-chains, a relatively large proportion of the CD8+ TCRγδ cells in the spleens of embryos and young chicks express only the α-chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved but there is a population of TCRγδ CD8αβ cells in the chicken that is not found in rodents. The monoclonal antibody EP72 recognizes the CD8α chain.

It is expressed on multipotent hematopoietic stem cells in bone marrow of mice with both the Ly-6.1 and Ly-6.2 allotypes. In the periphery, Ly-6A/E exhibits a pattern of expression which is based on differences between the two allotypes. Ly-6.1 strains (e.g., A, BALB/c, CBA, C3H/He, DBA/1, NZB) have few Ly-6A/E+ resting peripheral lymphocytes whereas Ly-6.2 strains (e.g., AKR, C57BL, C57BR, C57L, DBA/2, PL, SJL, SWR, 129) have relatively high numbers of Ly-6A/E+ lymphocytes. The expression of Ly-6A/E is dramatically upregulated in all strains upon cellular activation. Studies with the monoclonal antibody D7 suggest that Ly-6A/E may be involved in B and T lymphocyte responses and it appears that the antigen may be required for T-cell receptor-mediated T-cell activation.

Description: CD62L, also known as L-selectin, is a type I transmembrane glycoprotein with lectin-like and epidermal growth factor-like domains. It binds to sialylated oligosaccharide determinants on high endothelial venules (HEV) in peripheral lymph nodes. In the mouse, CD62L is expressed on most thymocytes and on peripheral leukocytes, including B and T lymphocytes, neutrophils, monocytes, and eosinophils. This member of the selectin family of cell adhesion molecules appears to be required for lymphocyte homing to peripheral lymph nodes and for leukocyte extravasation at sites of inflammation. Expression of CD62L on lymphocytes and neutrophils is rapidly down-regulated upon cell activation and the level of CD62L expression, along with other markers, distinguishes naive T cells from effector/memory T cells. The monoclonal antibody MEL-14 blocks in vitro binding of lymphocytes to peripheral lymph node HEV and inhibits in vivo lymphocyte extravasation into peripheral lymph nodes and the late stages of leukocyte rolling.

Description: The monoclonal antibody 20d5 reacts with an epitope common to the NKG2A, NKG2C and NKG2E isoforms of the NKG2 heterodimer, a member of the c-type lectin family of inhibitory receptors. Mouse NKG2 is a natural killer (NK) cell receptor for the non-classical MHC class I molecule Qa-1b. NKG2 is expressed on NK cells, lymphokine-activated killer (LAK) T cells, and some CD8+ memory T cells. Two-color immunofluorescent staining with the 20d5 antibody reveals two distinct NKG2+ phenotypes within the NK1.1+CD3- spleen cell population: NKG2hi (50%) and NKG2low (50%). With the exception of DBA/2J mice, NKG2 is expressed on NK1.1+ cells from all mouse strains tested (C57BL/6, BALB/c, 129/J, C3H.SW, AKR/J, SJL).
