Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

Description: The CD23 antigen is the low affinity IgE Fc receptor, which is a 45-49 kDa protein with 38 and 28 kDa fragments. It is expressed on most mature, conventional B cells (but not on peritoneal CD5+ B cells) and can also be found on the surface of T cells, macrophages and platelets. It is distinct from the high affinity IgE receptors found on basophils and mast cells which mediate allergic reactions. The low affinity receptors are thought to play a role in isotype specific immunoregulation. The regulation of CD23 surface expression appears to be integral with the complex IgE system which involves interactions of cells, cytokines, antibodies and regulatory factors. CD23 has been described as a “membrane bound cytokine," in that the soluble cleavage products of CD23 are themselves able to act as cytokines in vitro.

Description: CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

Description: CD33 is a 67 kDa type I transmembrane glycoprotein and a member of the sialoadhesin family of cell surface receptors. It is absent from pluripotent stem cells but appears on myelomonocytic precursors after CD34. It then continues to be expressed on both the myeloid and monocyte lineages. While it has been reported that CD33 can function as a sialic acid-dependent cell adhesion molecule, cells expressing CD33 require desialylation before they can bind cells bearing the appropriate sialoglycoconjugates. This suggests that inhibitory cis interactions may regulate or block any adhesion function.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The high affinity receptor complex is an α/β/γ heterotrimer with a Kd of 1.3 x 10-11 M. In mouse spleen, CD122 is expresed on ~30% of CD8+ cells and all NK cells but <1% of B cells and CD4+ T lymphocytes. In the thymus, its expression is confined to CD4-CD8+ single positive and CD4-CD8- double negative cells. Cytoplasmic regions of the IL-2R β chain are involved in IL-2-mediated cellular signaling and, via the interaction of IL-2 and its receptor complex, may be involved in the generation and differentiation of T lymphocytes. The monoclonal antibody 5H4 does not block IL-2 binding.

Description: Chicken Bu-1a, a product of the Bu-1a allele, is expressed on the cell surface as a disulfide-linked homodimer. In 10-week-old H.B15 and H.B2 chickens, Bu-1 is found on 85-90% of bursal cells, 2-8% of thymocytes, 15-27% of spleen cells, and 2-18% of peripheral blood cells. It is also expressed on subsets of macrophages and monocytes but not on granulocytes, erythrocytes, or thrombocytes. In chickens heterozygous for the Bu-1 alleles (Bu-1a/b), Bu-1a does not exhibit allelic exclusion. The monoclonal antibody 21-1A4 does not react with cells from CHA and H.B14ab strains by immunofluorescence.
