Description: DR5, also known as TRAIL-R2, TRICK2 and KILLER, is a widely expressed type I membrane member of the tumor necrosis (TNF) receptor family. Members of this family contain multiple cysteine-rich repeats in their extracellular domain and while the cytoplasmic region is less conserved, they do contain an intracellular 80 amino acid motif known as the ‘death domain’ (DD). DR5 is a receptor for the cytotoxic ligand TRAIL which induces apoptosis and it is the cytoplasmic DD that is responsible for transducing the death process. DR5 has also been shown to activate NF-κB.

Description: The CD117 (c-Kit) antigen is the cell-surface receptor for stem cell factor (SCF). Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: CD56, an isoform of the neural cell adhesion molecule (NCAM), is a heavily glycosylated type I transmembrane protein. On human hematopoietic cells its expression is restricted to NK cells and a subpopulation of T lymphocytes (known as large granular lymphocytes) which demonstrate natural killer activity. While it is clear that NCAM plays a role in homotypic adhesion of neuronal cells, the significance of CD56 expression on NK cells is not clear and remains controversial.

It is expressed on cytokine-stimulated endothelium, weakly on unactivated endothelium, and is absent on leukocytes. Like other selectins, CD62E is an adhesion molecule that contributes to the initial attachment and rolling of leukocytes on endothelial surfaces, a prerequisite for leukocyte extravasation into tissues. CD62P is a 140 kDa membrane glycoprotein formerly known as platelet activation-dependent granule membrane protein (PADGEM) or GMP-140. It is expressed on platelets, megakaryocytes, and epithelium. Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. Expression of CD25 on activated T lymphocytes is transitory and endogenously regulated. CD25 is expressed on precursor B cells in bone marrow. Its expression is initiated by functional rearrangement and expression of IgM heavy chain genes and is down-regulated when immature B cells mature and express IgD. It is expressed at a higher level on CD4+CD8+ thymocytes. It is also expressed on cultured epidermal Langerhans cells. The biochemical features of murine CD25 have been characterized in detail. The 3C7 monoclonal antibody reacts with an epitope of CD25 which is distinct from that recognized by clone 7D4. 3C7, used in combination with 7D4 in culture, results in higher levels of inhibition of proliferation driven by IL-2 and generation of alloreactive CTL than either of these monoclonal antibodies alone.

Description: The monoclonal antibody TCR-2 precipitates a CD3-associated heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 34 kDa and 29 kDa. In the chicken, two distinct subpopulations of αβ T cells appear in the thymus subsequent to the appearance of γδ T cells. These subpopulations, originally denoted as TCR2 and TCR3, arise sequentially in the thymus during ontogeny and are now known to represent two distinct Vβ families, Vβ1 and Vβ2, respectively. The TCR-2 monoclonal antibody reacts with approximately 40% of thymocytes, 40-50% of blood mononuclear cells, and 40% of splenocytes in the chicken. Two-color immunofluorescence has revealed that most of the TCR2+ thymocytes express both CD4 and CD8 antigens. The TCR2+ cells in blood were found to express either CD4 (74 ± 2%) or CD8 (26 ± 4%). TCR2+ cells in the spleen also express either CD4 (37 ± 1%) or CD8 (64 ± 4%). Surprisingly, a relatively large subpopulation of CD8+ cells in the spleen are negative for TCR2. This observation led to the demonstration that 71 ± 6% of the TCRγδ+ cells in the spleen express CD8.
