Description: CD152, also known as CTLA-4, is homologous to the costimulatory molecule CD28. Like CD28, the CD152 molecule binds the family of B7 counter-receptors (CD80 and CD86) on antigen-presenting cells. CTLA-4 is not expressed on resting T lymphocytes but is induced 2-3 days following T cell stimulation via the T cell receptor (TCR) complex. Whereas CD28 provides a positive costimulatory signal, CD152 is a negative regulator of T-cell activation, possibly by inhibiting tyrosine kinase signaling through the TCR through its association with tyrosine phosphatases such as SHP-2.

Description: Murine CD72.1 (Lyb-2.1), a type II integral membrane glycoprotein and a member of the C-lectin family of cell surface receptors, is a differentiation antigen of B cells and is found in mouse strains expressing the Lyb-2.1 allotype. CD72.1 is the ligand of CD5 which is distributed on all T cells and a small number of B cells. The 10.1.D2 monoclonal antibody blocks binding of CD5 to CD72 on the B cell surface which leads to inhibition of the positive signal resulting from CD5/CD72 pairing. However, 10.1.D2 can itself trigger a positive signal by binding CD72.

Description: CD24 is expressed in B cells until their terminal differentiation into plasma cells. It has been used widely as a marker to fractionate different B-lineage differentiation stages. CD24 has been found on a variety of other hematopoietic and neural cell types and is now classified as a costimulatory molecule. The CD24 molecule, also known as HSA, is composed mostly of carbohydrate with a protein core of only 31-35 amino acids in the human and 27 amino acids in the mouse. CD24 is now included in the large group of phosphatidylinositol-anchored membrane proteins. Studies with purified CD24 applied to latex beads have demonstrated a self-binding property suggesting that the molecule is capable of promoting homotypic aggregation, binding, and cell adhesion. This is supported by earlier observations of the blockade of B cell lymphoblast aggregation with an anti-CD24 monoclonal antibody. Recent studies of murine CD24 have assigned a role for CD24 as a ligand for P-selectin, a calcium-dependent lectin.

Description: CD22, also known as Lyb-8, is a heterodimeric transmembrane glycoprotein and a member of a structurally related group of immunoglobulin (Ig) superfamily domain-containing proteins called the sialoadhesin family. It is detected in the cytoplasm early in B cell development (late pro-B cell stage), appears on the cell surface concomitant with IgD, and is found on most IgM+IgD+ mature B lymphocytes. Expression is lost with terminal differentiation of B cells and is absent on plasma cells. Activation of B cells via cross-linking of surface Ig increases CD22 expression. CD22 associates with the B cell receptor (BCR) complex and mediates intercellular adhesion. Its intracellular domain is phosphorylated after antigen receptor cross-linking and is involved in negative regulation of B cell activation.

Description: Chicken CD45 is a transmembrane glycoprotein expressed on all leukocytes. B lineage cells in the bursa of Fabricius express the 215 kDa isoform while T lineage cells bear the 190 kDa variant. These high molecular weight molecules have intrinsic phosphotyrosine phosphatase activity characteristic of mammalian CD45. Levels of CD45 expression detected by the monoclonal antibody LT40 in the avian thymus are heterogeneous with approximately 90% of thymocytes expressing fourfold higher levels of surface CD45 (CD45hi) than do the remaining 10% (CD45low) of thymocytes. The CD45low population contains exclusively thymocytes with the CD3-CD4-CD8low phenotype characteristic of the immediate precursors to the CD3-CD4+CD8+ thymic population which are CD45hi. This shift from low to high levels of surface CD45 expression is concomitant with the transition from CD4-CD8low to CD4+CD8+ and before the expression of CD3. The ability of the monoclonal antibody LT40 to discriminate this CD45low subpopulation provides a suitable marker for the CD3-CD4-CD8low immediate precursors to the CD3-CD4+CD8+ thymocytes.

Description: CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion. CD107a is expressed by activated T cells, macrophages, dendritic cells, activated platelets, tonsillar epithelium, and some tumor cell lines, including U937 and KG1a. It is also a widely expressed intracellular antigen. LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases.
