It is constitutively expressed on the surface of endothelial cells and concentrated at the junction between them. It is also weakly expressed on many peripheral lymphoid cells and platelets. CD31 interacts homotypically in cell adhesion assays. The monoclonal antibody 390 inhibits the aggregation of L cells transfected with a variant form of CD31.

Description: Interleukin-25 (IL-25), also known as interleukin-17E (IL-17E), is a 17 kDa protein and member of the IL-17 cytokine family. This cytokine is secreted by a variety of cells including type 2 helper T cells (Th2), bone marrow-derived mast cells, and alveolar macrophages. On target cells, IL-25 can induce the production of several type 2 cytokines including IL-4, IL-5, and IL-13 and implicates a role for this cytokine in type 2 immune response. Studies have linked this cytokine in promoting airway hyperreactivity, limiting pro-inflammatory cytokine production and chronic intestinal inflammation, and providing protective immunity following intestinal helminth infection.

Description: The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes, which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 34-1-2S binds to a common determinant in the α3 domains of H-2Kd and H-2Dd in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Kb.

Description: The CD117 (c-Kit) antigen is the cell-surface receptor for stem cell factor (SCF). Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: CD107a, also known as lysosomal-associated membrane protein 1 (LAMP-1), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains, and is a ligand for E-selectin-mediated cell adhesion. CD107a is expressed by activated T cells, macrophages, dendritic cells, activated platelets, tonsillar epithelium, and some tumor cell lines, including U937 and KG1a. It is also a widely expressed intracellular antigen. LAMP-1 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

Description: Ly-6A/E, also known as Sca-1, is a member of the Ly-6 multigene family of type V glycophosphatidylinositol-anchored cell surface proteins. It is expressed on multipotent hematopoietic stem cells in bone marrow of mice with both the Ly-6.1 and Ly-6.2 allotypes. In the periphery, Ly-6A/E exhibits a pattern of expression which is based on differences between the two allotypes. Ly-6.1 strains (e.g., A, BALB/c, CBA, C3H/He, DBA/1, NZB) have few Ly-6A/E+ resting peripheral lymphocytes whereas Ly-6.2 strains (e.g., AKR, C57BL, C57BR, C57L, DBA/2, PL, SJL, SWR, 129) have relatively high numbers of Ly-6A/E+ lymphocytes. The expression of Ly-6A/E is dramatically upregulated in all strains upon cellular activation. Studies with the monoclonal antibody D7 suggest that Ly-6A/E may be involved in B and T lymphocyte responses and it appears that the antigen may be required for T-cell receptor-mediated T-cell activation.

Description: The monoclonal antibody 5H10.27 reacts with the α5 chain (VLA-5) of the CD49e/CD29 heterodimeric fibronectin receptor. It is expressed on thymocytes, activated T cells, mast cells, and a variety of mouse cell lines. Soluble 5H10.27 antibody inhibits VLA-5-mediated functions in vitro. Immobilized 5H10.27 has also been demonstrated to costimulate the proliferative response of CD8+ T cells to plate-bound anti-CD3 monoclonal antibody.
