Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. Expression of CD25 on activated T lymphocytes is transitory and endogenously regulated. CD25 is expressed on precursor B cells in bone marrow. Its expression is initiated by functional rearrangement and expression of IgM heavy chain genes and is down-regulated when immature B cells mature and express IgD. It is expressed at a higher level on CD4+CD8+ thymocytes. It is also expressed on cultured epidermal Langerhans cells. The biochemical features of murine CD25 have been characterized in detail. The 3C7 monoclonal antibody reacts with an epitope of CD25 which is distinct from that recognized by clone 7D4. 3C7, used in combination with 7D4 in culture, results in higher levels of inhibition of proliferation driven by IL-2 and generation of alloreactive CTL than either of these monoclonal antibodies alone.

Description: Protein tyrosine residues are phosphorylated as a result of intracellular protein kinase activation (e.g., via growth factors) during normal growth and development and in oncogenesis. The most abundant population of target proteins for tyrosine phosphorylation is cell surface glycoproteins. Antibodies to phosphotyrosine enable the detection, isolation, and characterization of proteins containing phosphotyrosine. The monoclonal antibody PY20 prevents internalization of activated receptors (e.g., EGFR) when microinjected into cells. The affinity of PY20 for phosphotyrosine is approximately 10-6 to 10-7 M. PY20 binding to phosphorylated tyrosines can be inhibited by free phosphotyrosine and phenylphosphate but not by phosphoserine, phosphothreonine, or free phosphate.

Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Immobilized 145-2C11 can activate both normal T lymphocytes and cloned T cell lines. Under certain conditions, T cell activation by 145-2C11 may result in apoptotic cell death.

Description: CD24, also known as heat stable antigen, is a phosphatidylinositol-linked cell surface glycoprotein that is expressed at all stages of B cell development and on most thymocytes. The absence of expression from mature T cells is closely associated with their maturation from CD4+CD8+CD24+ thymocytes to either CD4+CD8-CD24- or CD4-CD8+CD24- T cells. CD24 is also present on mouse granulocytes, monocytes, Langerhans cells and erythrocytes. Its ligand is P-selectin (CD62P). CD24 may play a role in regulation of B cell proliferation and differentiation, and it has been proposed that heat stable antigen promotes homotypic adhesion between B lymphocytes.
