Description: Murine CD72.1 (Lyb-2.1), a type II integral membrane glycoprotein and a member of the C-lectin family of cell surface receptors, is a differentiation antigen of B cells and is found in mouse strains expressing the Lyb-2.1 allotype. CD72.1 is the ligand of CD5 which is distributed on all T cells and a small number of B cells. The 10.1.D2 monoclonal antibody blocks binding of CD5 to CD72 on the B cell surface which leads to inhibition of the positive signal resulting from CD5/CD72 pairing. However, 10.1.D2 can itself trigger a positive signal by binding CD72.

It is expressed on bone marrow cells, monocytes/macrophages, neutrophils, endothelial cells, and T-cell subsets. Mice with the Ly-6.2 allotype (e.g., AKR, C57BL, C57BR, C57L, DBA/2, PL, SJL, SWR, 129) have subsets of CD4+Ly-6C+ and CD8+Ly-6C+ cells while Ly-6.1 strains (e.g., A, BALB/c, CBA, C3H/He, DBA/1, NZB) have only CD8+Ly-6C+ lymphocytes. Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.
