Description: CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. It also reacts with certain activated T cells as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells. Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Description: CD107b, also known as lysosomal-associated membrane protein 2 (LAMP-2), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains and is a ligand for E-selectin-mediated cell adhesion. CD107b is expressed in lysosomal/endosomal membranes and on the surface of activated platelets and some tumor cell lines including U937 and KG1a. LAMP-2 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.
