Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description: Chicken CD45 is a transmembrane glycoprotein expressed on all leukocytes. B lineage cells in the bursa of Fabricius express the 215 kDa isoform while T lineage cells bear the 190 kDa variant. These high molecular weight molecules have intrinsic phosphotyrosine phosphatase activity characteristic of mammalian CD45. Levels of CD45 expression detected by the monoclonal antibody LT40 in the avian thymus are heterogeneous with approximately 90% of thymocytes expressing fourfold higher levels of surface CD45 (CD45hi) than do the remaining 10% (CD45low) of thymocytes. The CD45low population contains exclusively thymocytes with the CD3-CD4-CD8low phenotype characteristic of the immediate precursors to the CD3-CD4+CD8+ thymic population which are CD45hi. This shift from low to high levels of surface CD45 expression is concomitant with the transition from CD4-CD8low to CD4+CD8+ and before the expression of CD3. The ability of the monoclonal antibody LT40 to discriminate this CD45low subpopulation provides a suitable marker for the CD3-CD4-CD8low immediate precursors to the CD3-CD4+CD8+ thymocytes.

Description: The lymphocyte Fcγ receptors recognize the Fc portion of IgG, presented either as immune complexes or as free antibody. The different classes of receptors are distinct because of varying size, tissue distribution and affinity for IgGs. The Fc type II receptor is expressed on a wide variety of cells including B cells, hematopoietic cells, monocyte/macrophages, neutrophils, platelets, Langerhans cells, eosinophils, basophils, trophoblasts, and endothelial cells of the placenta. The Fcγ type III receptors are higher affinity than the type II and are expressed on macrophages, NK cells and neutrophils. Both types of receptors can be expressed on the same cell and in varying ratios. The receptors are constitutively expressed, although cytokines and lymphokines can modulate their expression. Besides identifying FcγR+ cells, monoclonal antibodies to the FcγII/III receptor have been used to block Fc receptor binding of IgG, Fc-mediated signal transduction and effector functions, clearance of immune complexes and to attenuate infection by organisms dependent on FcγR for parasitic invasion. Description: CD1 molecules, like MHC I and II, play an important role in the immune system by presenting lipid, glycolipid and lipopeptide antigen to T and NKT cells. The chicken CD1 gene is located in the chicken MHC B locus and has an important implication for the primordial MHC gene evolution. Two proteins have been identified - CD1.1 and CD1.2. Analysis of RNA from blood cells showed that both genes are expressed in Bu-1+ cells and CD8α+ cells. CD1.2 is expressed in TCRγδ cells but is nearly undetectable in TCRαβ. The monoclonal antibody CB3 antibody can identify cells of the B cell lineage and recognizes CD1.1 but not CD1.2 on the surface of transfected mammalian cells.

Description: CD95, also known as Fas and APO-1, is a 45 kDa type I transmembrane glycoprotein and a member of the tumor necrosis factor receptor superfamily. It is expressed by activated lymphocytes, monocytes, neutrophils, fibroblasts, and cell lines. Fas ligand binding to CD95 induces apoptosis in activated mature lymphocytes thereby playing a role in maintaining peripheral tolerance. Crosslinking of CD95 by the monoclonal antibodies DX2 and DX3 delivers an apoptotic signal to Fas-sensitive cells indicating that these monoclonal antibodies recognize a functional epitope of CD95.
