Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Description: CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.

Description: CD106, also known as INCAM-110, is a 110 kDa vascular adhesion cell adhesion molecule-1 (VCAM-1) that is member of the immunoglobulin superfamily. CD106 is expressed predominantly on cytokine-activated vascular endothelium but has also been identified on interfollicular dendritic cells, some macrophages, and bone marrow stromal cells. Endothelial CD106 binds the integrins α4β1 (CD49d/CD29, VLA-4) and α4β7 and contributes to extravasation of lymphocytes, monocytes, basophils, and eosinophils (but not neutrophils) from blood vessels, particularly at sites of inflammation. Unlike the β2 integrins, the CD106-VLA-4 interaction can mediate both the initial tethering and rolling of lymphocytes on endothelium as well as their subsequent arrest and firm adhesion. CD106 expressed on non-vascular tissues has been implicated in the interaction of hematopoietic progenitors with bone marrow stromal cells, B cell binding to follicular dendritic cells, costimulation of T cells, and embryonic development. The monoclonal antibody 1.G11B1 inhibits in vitro binding of lymphocytes and monocytes to VCAM-1 on stimulated endothelium.
