Description: This antibody recognizes a protein of 205 kDa-220 kDa, identified as CD45RA. CD45RA is isoforms of the human leukocyte common antigen (CD45). Human CD45 contains three exons which encode peptide segments designated A, B and C, respectively. The differential splicing of the exons generates at least five isoforms, ABC, AB, BC, B and O. This antibody reacts with ABC and BC isoforms. CD45RA is expressed on 40-50% of peripheral CD4 T-cells, 50% of peripheral CD8 T-cells, B-cells, and leukemic B-cell lines. T-cells expressing CD45RA are naive or virgin T-cells. T-cells expressing CD45RO are memory T-cells. CD45RA and CD45RO define complementary, predominantly non-overlapping populations of resting peripheral T-cells. This MAb is useful in study on the subpopulation of CD4 or CD8 T-cells. It can especially be used to differentiate T-cell lymphomas (CD45RO ve) from B cell lymphomas (CD45RA ve).

Description: Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms. Most pituitary adenomas and prolactinomas readily express chromogranin.

CF®405S is a blue fluorescent dye (Ex/Em 404/431 nm) with superior brightness compared to other blue dyes; it is also compatible with super-resolution imaging by SIM. Description: This MAb specifically precipitates heterogeneous material of high MW, identified as perlecan, a major heparan-sulfate proteoglycan (HSPG) within all basement membranes and cell surfaces. It does not cross-react with laminin, fibronectin, or dermatran sulfate proteoglycan. Because of perlecan s strategic location and ability to store and protect growth factors, it has been strongly implicated in the control of tumor cell growth and metastatic behavior. Perlecan possesses angiogenic and growth-promoting attributes primarily by acting as a co-receptor for basic fibroblast growth factor (FGF-2). Suppression of perlecan causes substantial inhibition of neoplastic growth and neovascularization. Thus, perlecan is a potent inducer of neoplasm growth and angiogenesis in vivo and therapeutic interventions targeting this key modulator of tumor progression may improve neoplastic treatment.

Description: This antibody recognizes actin of skeletal, cardiac, and smooth muscle cells. It is not reactive with other mesenchymal cells except for myoepithelium. Actin can be resolved on the basis of its isoelectric points into three distinctive components: alpha, beta, and gamma in order of increasing isoelectric point. Anti-muscle specific actin recognizes alpha and gamma isotypes of all muscle groups. Non-muscle cells such as vascular endothelial cells and connective tissues are non-reactive. Also, neoplastic cells of non-muscle-derived tissue such as carcinomas, melanomas, and lymphomas are negative.It stains tumors of smooth muscle (leiomyomas and leiomyosarcomas) as well as skeletal muscle (rhabdomyomas and rhabdomyosarcomas).

Description: This antibody recognizes a protein of ~55 kDa, identified as SOX10. This MAb is highly specific and does not cross-react with other members of the SOX-family. SOX genes comprise a family of genes that are related to the mammalian sex-determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. SOX-10 is a sensitive marker of melanoma, including conventional, spindled, and desmoplastic subtypes. It is expressed by metastatic melanomas and nodal capsular nevus in sentinel lymph nodes, but not by other lymph node components such as dendritic cells, which usually express S100 protein. Commonly used melanoma markers, such as anti-HMB-45 and anti-Melan-A, are poorly expressed in desmoplastic melanomas while SOX-10 is moderately to strongly expressed in desmoplastic melanomas. SOX-10 is considered as a very reliable marker for recognizing residual desmoplastic melanomas. In normal tissues, it is expressed in Schwann cells, melanocytes, and myoepithelial cells of salivary, bronchial and mammary glands. SOX-10 expression is also observed in mast cells.

Description: This antibody recognizes a cell surface glycoprotein of 80-95 kDa (CD44) on lymphocytes, monocytes, and granulocytes (Leucocyte Typing Workshop V). Its epitope is resistant to digestion by trypsin and chymotrypsin. The CD44 family of glycoproteins exists in a number of variant isoforms, the most common being the standard 85-95 kDa or hematopoietic variant (CD44s). Higher molecular weight isoforms are described in epithelial cells (CD44v), which are believed to function in intercellular adhesion and stromal binding. CD44 immunostaining is commonly used for the discrimination of urothelial transitional cell carcinoma in-situ from non-neoplastic changes in the urothelium.
