Description: Interleukin-33 (IL-33; HF-NEV; IL-1F11), a member of the IL-1 family of cytokines, is expressed by many cell types following pro-inflammatory stimulation and is thought to be released upon cell lysis. IL-33 binds to and signals through ST2 (IL-1R1) and its stimulation recruits MYD88, IRAK, IRAK4 and TRAF6, followed by phosphorylation of ERK1 (MAPK3)/ERK2 (MAPK1), p38 (MAPK14) and JNK. The ability of IL-33 to target numerous immune cell types, like Th2-like cells, mast cells and B1 cells, and to induce cytokine and chemokine production underlines its potential in influencing the outcome of a wide range of diseases, such as arthritis, asthma, atopic allergy & anaphylaxis, cardiovascular disease/atherosclerosis, nervous system diseases and sepsis. Anti-IL-33, mAb (recombinant) (blocking) (Bondy-1-1) is an antibody developed by antibody phage display technology using a human naive antibody gene library. These libraries consist of scFv (single chain fragment variable) composed of VH (variable domain of the human immunoglobulin heavy chain) and VL (variable domain of the human immunoglobulin light chain) connected by a polypeptide linker. The antibody fragments are displayed on the surface of filamentous bacteriophage (M13). This scFv was selected by affinity selection on antigen in a process termed panning. Multiple rounds of panning are performed to enrich for antigen-specific scFv-phage. Monoclonal antibodies are subsequently identified by screening after each round of selection. The selected monoclonal scFv is cloned into an appropriate vector containing a Fc portion of interest and then produced in mammalian cells to generate an IgG like scFv-Fc fusion protein.

Description: Bmf (Bcl-2-modifying factor) belongs to the Bcl-2 protein family. Bcl-2 family members form hetero- or homo-dimers and act as anti- or pro-apoptotic regulators that are involved in a wide variety of cellular activities. Bmf contains a single Bcl-2 homology domain 3 (BH3), and has been shown to interact with other Bcl-2 proteins and functions as an apoptotic activator. Two different start sites can give rise to two proteins of 25 or 30 kDa. It is found to be sequestered to myosin V motors by its association with dynein light chain 2 (DLC2), which may be important for sensing intracellular damage and triggering apoptosis.

Description: Angiopoietin-1 (Ang-1) and Angiopoietin-2 (Ang-2) are closely related secreted ligands which bind with similar affinity to Tie-2. Tie-2 and angiopoietins have been shown to play critical roles in embryogenic angiogenesis and in maintaining the integrity of the adult vasculature. Ang-1 activates Tie-2 signaling on endothelial cells to promote chemotaxis, cell survival, cell sprouting, vessel growth and stabilization. Ang-2 has been identified as a secreted protein ligand of Tie-2 and has alternatively been reported to be an antagonist for Ang-1 induced Tie-2 signaling as well as an agonist for Tie-2 signaling, depending on the cell context.

Description: Human CD155 (Polio Virus Receptor, PVR, Necl-5) is a 70kd type I Ig superfamily molecule. It is involved in formation of intracellular junctions between epithelial cells. Its ligands include CD226 (DNAM-1), and CD96 (TACTILE). CD155 expression by tumor has been shown to be upregulated by nitric oxide. A soluble version of CD155 has been shown to exist. High CD155 expression has recently been exploited to use engineered poliovirus to treat glioblastoma.

Description: Interleukin-8 (IL-8) was originally discovered as a neutrophil chemotactic and activating factor and is a member of the alpha (CXC) subfamily of chemokines (including also platelet factor 4, GRO, IP-10, etc.). Many cell types, including monocyte/macrophages, T cells, neutrophils, fibroblasts, endothelial cells, keratinocytes, hepatocytes, chondrocytes and various tumor cell lines, produce IL-8 in response to a wide variety of proinflammatory stimuli such as exposure to IL-1, TNF, LPS and viruses. IL-8 has a wide range of other proinflammatory effects. It is a potent chemoattractant for neutrophils and causes degranulation of neutrophil specific granules and azurophilic granules. IL-8 induces expression of the cell adhesion molecules CD11/CD18 and enhances the adherence of neutrophils to endothelial cells and subendothelial matrix proteins. Besides neutrophils, IL-8 is also chemotactic for basophils, T cells and eosinophils. IL-8 has been reported to be a co-mitogen for keratinocytes and was also shown to be an autocrine growth factor for melanoma cells. IL-8 was also reported to be angiogenic both in vivo and in vitro.

Description: Human CD45 is expressed on all cells of hematopoietic origin, except erythrocytes. CD45 is a transmembrane tyrosine phosphatase which can exist in at least nine different isoforms resulting from tissue-specific alternative RNA splicing of exons 4-7 of a single gene coding for the various N-terminal peptide segments. The CD45RO isoform contains none of the alternatively spliced exons and is present on most thymocytes, a subpopulation of resting (memory) T cells within both CD4 and CD8 subsets as well as on monocytes and granulocytes. CD45RO plays an essential role in lymphocyte activation.

Description: Human CD28 is an important costimulatory molecule found on all CD4+ T cells and on about half of the CD8+ T cells. T cell activities attributed to CD28 include prevention of anergy, induction of cytokine gene transcription, stabilization of cytokine mRNA and activation of CD8+ cytotoxic T lymphocytes. The ligands for CD28 identified as CD80 (B7-1) and CD86 (B7-2) are immunglobulin superfamily monomeric transmembrane glycoproteins of 60kDa and 80kDa respectively. Anti-CD28 antibodies have been shown to have a strong adjuvant effect when conjugated to an immunogen. Monovalency of the CD28 receptor for its natural ligands is essential to provide costimulation without inducing responses in the absense of TCR engagement.

Description: TRAIL-R1, -R2, -R3 and -R4 are receptors for the cytotoxic ligand TRAIL. When signaling through TRAIL-R1 and -R2, the adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. They promote the activation of NF-kappaB. TRAIL-R3 lacks a cytoplasmic death domain, and TRAIL-R4 contains a truncated death domain, and hence are not capable of inducing apoptosis. They protect cells against TRAIL mediated apoptosis by competing with TRAIL-R1 and R2 for binding to the ligand.

Description: Neuregulin-4 (Nrg4) belongs to a small family of EGF-like (EGFL) domain-containing proteins that are synthesized as transmembrane precursors and undergo proteolytic cleavage. The EGF-like domain (aa 5-46) of Nrg4 (aa 1-53) directly binds to the receptors ErbB3 and 4. Nrg4 is a cold induced adipokine, highly expressed in adipose tissues and enriched in brown fat. It is increased during brown adipocyte differentiation and reduced in rodent and human obesity. It promotes neurite outgrowth and protects against diet-induced insulin resistance and hepatic steatosis through attenuating hepatic lipogenic signaling. This hepatic effect of Nrg4 is mediated by ErbB3 and ErbB4 signaling that negatively regulates de novo lipogenesis mediated by LXR and SREBP1c. This effect of Nrg4 on fatty liver and insulin resistance could lead to the development of Nrg4 as an effective therapeutic biological for the treatment of NAFLD and type 2 diabetes. GST-Nrg4 (aa 1-53) recombinant protein has been shown to mimic the effect of endogenous secreted Nrg4 on liver lipogenesis.
