Description: Musashi2 (MSI2) is an RNA-binding protein that is highly expressed in precursor cells in the ventricular and subventricular zones of the developing mammalian CNS. Like the related MSI1, MSI2 has been suggested to be involved stem cell production and maintenance. MSI2 is the predominant MSI protein in hematopoietic stem cells, and its knockdown leads to reduced engraftment and depletion in vivo. Expression levels of MSI2 are elevated in myeloid leukemia cells lines, and MSI2 appears to cooperate with BCR-ABL1 to induce an aggressive leukemia; the level of MSI2 directly correlates with decreased survival in patients. MSI2 negatively regulates the asymmetric cell fate determinant NUMB, suggesting that this signaling pathway may provide future targets for future therapies.

Description: NALP1 belongs to a family of cytoplasmic proteins that have been implicated in cell responses to apoptotic and inflammatory stimuli. It contains a caspase recruitment domain (CARD) in addition to a NACHT domain and leucine rich repeat (LRR). This protein interacts strongly with caspase 2 and weakly with caspase 9, as well as with Apaf-1. Transient overexpression of this gene in cultured cells was sufficient to induce apoptosis. NALP1 mRNA and protein expression levels in myeloid leukemia cells are increased following CREB (cAMP-response element binding protein) activation, suggesting that NALP1 may contribute to modulate the response of these cells to pro-inflammatory stimuli. Multiple alternatively spliced transcript variants encoding distinct isoforms have been found for NALP1.

Description: JAKMIP1, also known as Marlin-1, is a highly-conserved vertebrate-specific microtubule-associated protein that plays a role in JAK1 signaling and regulate microtubule cytoskeleton rearrangements. JAKMIP1 is expressed predominantly in neuronal and lymphoid cells and colocalizes with microtubules. Multiple isoforms of JAKMIP1 are known to exist: isoforms 1 and 5 are also expressed in liver, lung and skeletal muscle, while isoforms 2-4 are specifically expressed in brain and retina. JAKMIP1 participates in polarized secretion in lymphocytes and it has been suggested to be a novel effector memory gene restraining T cell-mediated cytotoxicity. Dysregulation of JAKMIP1 gene expression is associated with genetic conditions linked to autism in humans.

Description: HSF1, or heat shock factor 1, belongs to a family of Heat Shock transcription factors that activate the transcription of genes encoding products required for protein folding, processing, targeting, degradation, and function (2). The up-regulation of HSP (heat shock proteins) expression by stressors is achieved at the level of transcription through a heat shock element (HSE) and a transcription factor (HSF) (3, 4, 5). Most HSFs have highly conserved amino acid sequences. On all HSFs there is a DNA binding domain at the N-terminus. Hydrophobic repeats located adjacent to this binding domain are essential for the formation of active trimers. Towards the C-terminal region another short hydrophobic repeat exists, and is thought to be necessary for suppression of trimerization (6). There are two main heat shock factors, 1 and 2. Mouse HSF1 exists as two isoforms, however in higher eukaryotes HSF1 is found in a diffuse cytoplasmic and nuclear distribution in un-stressed cells. Once exposed to a multitude of stressors, it localizes to discrete nuclear granules within seconds. As it recovers from stress, HSF1 dissipates from these granules to a diffuse nuceloplasmic distribution. HSF2 on the other hand is similar to mouse HSF1, as it exists as two isoforms, the alpha form being more transcriptionally active than the smaller beta form (7, 8). Various experiments have suggested that HFS2 may have roles in differentiation and development (9, 10, 11). Description: GOLPH1, also known as GCP60, was initially identified as a Golgi protein that can interact with the integral membrane protein giantin and is thought to be involved in the maintenance of the Golgi structure. GOLPH1 has also been shown to interact with other Golgi proteins such as Golgin-160, a Golgi protein that can be cleaved by caspases-2, -3, and -7, leading to the nuclear localization of Golgin-160. Description: The mediator complex is a multi-protein transcriptional co-activator that is expressed ubiquitously in eukaryotes from yeast to mammals and is required for induction of RNA polymerase II (pol II) transcription by DNA binding transcription factor. One of the proteins in this complex is MED28, also known as Magicin. MED28 is expressed in many cell lines and tissues. It has been shown that a down-regulation of MED28 expression in NIH3T3 cells results in a significant induction of several genes associated with smooth muscle cell differentiation and conversely its over-expression represses expression of SMC genes. MED28 can also form a ternary complex with Grb2 and Merlin, the neurofibromatosis 2 tumor suppressor protein, indicating that MED28 may play a role in Merlin's tumor suppressive activity.
