Description: CD43, a member of the sialomucin family of cell surface receptors, is a 95-135 kDa type I transmembrane glycoprotein whose variability in molecular weight is dependent upon the nature of the extracellular O-glycans. It is the major sialoglycoprotein on thymocytes, T cells, and neutrophils. It is also present on activated B cells, plasma cells, NK cells, granulocytes, monocytes, macrophages, platelets, and bone marrow hematopoietic stem cells. CD43, which binds to ICAM-1 (CD54), appears to function as an anti-adhesion molecule, inhibiting T cell interactions including T cell killing, and by increasing the threshold for T cell activation.

Description: CD5, also known as Lyt-1, is a monomeric type I transmembrane glycoprotein expressed on thymocytes, T lymphocytes, and a subset of B lymphocytes, but not on natural killer (NK) cells. It has been identified as the major ligand of the B-cell antigen CD72. The frequency of CD5+ B cells exhibits strain-dependent variation, and the phenotypic, anatomical, functional, developmental, and pathological characteristics of the CD5+ B cells suggest that they may represent a distinct lineage, known as B-1 cells. Binding of CD5 on the T cell surface can augment alloantigen- or mitogen-induced lymphocyte proliferation and induces increased cytosolic free calcium, IL-2 secretion, and IL-2R expression. It has been proposed that CD5 negatively regulates signal transduction mediated by the T-cell and B-cell receptors.

Description: CD40 is a type I cell surface protein belonging to the tumor necrosis factor superfamily of cell surface receptors. In mice it is expressed on B lineage cells, follicular dendritic cells, thymic epithelium, and interdigitating cells in the T-cell zone of secondary lymphoid organs. CD40 first becomes detectable on a subset of small pre-B II cells in bone marrow with the levels of CD40 expression increasing thereafter during B cell maturation. Immature B cells (IgM+IgDloB220lo) express intermediate levels of CD40, whereas mature B cells (IgM+IgDhiB220hi) express high levels. CD40 has a central role in B cell growth and differentiation, and signaling through CD40 in combination with IL-4 reportedly induces immunoglobulin isotype switching and secretion of IgE. The agonistic 1C10 antibody closely resembles gp39/CD40 ligand in its ability to stimulate proliferation of small, resting B lymphocytes in the absence of other cofactors.

Description: The “classical” MHC Class I molecules are histocompatibility antigens encoded by the H-2 gene complex and consist of heterodimers of highly polymorphic α chains noncovalently associated with the invariant β2-microglobulin. These antigens are expressed on most nucleated cells but expression varies on different cell types. MHC Class I molecules present endogenously synthesized peptides to CD8+ T lymphocytes which are usually cytotoxic T cells. MHC Class I antigens expressed on thymic epithelial cells regulate the positive and negative selection of CD8+ T cells during T cell ontogeny. The monoclonal antibody 28-14-8 binds to the α3 domain of H-2Db in the presence or absence of β2 microglobulin. It cross reacts with the α3 domain of H-2Ld, but not Kd or Dd, and with H-2Dq and/or Lq. The antibody has been shown to block H-2Ld-specific and H-2Ld-restricted antigen-specific lysis of target cells by cytotoxic T lymphocytes but it does not block recognition of H-2Ld-positive target cells by Ly-6G2-positive NK cells.

Description: CD74 represents a type II integral membrane protein known as MHC class II-associated invariant chain. It is moderately expressed on B cells and weakly on monocytes and macrophages. CD74 is also found on a subset of activated T cells, Langerhans cells, dendritic cells, and some epithelial cells. CD74 may play a role in antigen processing and presentation through intracellular transport and expression of class II molecules on the cell surface.
