Description: CD45R, also known as B220, a member of the protein tyrosine phosphate family and a major cell surface glycoprotein, represents a restricted form of the CD45 family which primarily recognizes only cells of B-lineage from pro-B cell through mature B lymphocytes and prior to the availability of anti-CD19 monoclonal antibodies was commonly used as a pan B-cell marker. It also reacts with certain activated T cells as well as non-MHC-restricted lytically active lymphokine-activated killer (LAK) cells. In vivo administration of RA3-6B2 has been shown to affect differentiation of both T and B cells in normal mice and reduce the level of anti-DNA antibodies and lymphadenopathies in MRL/lpr mice.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosphorylate the Src family of protein kinases. This activity is now known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685, and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The former arises as a result of alternative splicing of different exons. The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. The monoclonal antibody A20 recognizes all mouse leukocytes expressing the CD45.1 allotype.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow.
