Description: CD69, also known as very early activation (VEA) antigen, is a disulfide-linked transmembrane homodimer whose differentially glycosylated subunits range from 35-39 kDa. It is a C-type lectin most closely related to the NKR-P1 and Ly-49 NK cell-activation molecules. CD69 is widely expressed on hematopoietic cells, including lymphocytes, neutrophils and eosinophils. Although not detectable on resting lymphocytes, its expression is rapidly upregulated upon activation of T, B and NK cells, and neutrophils. Constitutive expression of CD69 on subsets of thymocytes suggests that it may be involved in regulation of developmental events in addition to its role in activation of a variety of hematopoietic cells. The monoclonal antibody H1.2F3 augments PMA-induced T-cell proliferation and induces redirected lysis of Fc receptor-bearing target cells by NK cells.

Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

Description: In the chicken, the CD8 molecule is present in two forms - (i) a homodimer of two α chains and (ii) a heterodimer of an α chain and a β chain. While the vast majority of CD8+ cells in the thymus, spleen, and blood of adult chickens express both CD8α- and CD8β-chains, a relatively large proportion of the CD8+ TCRγδ cells in the spleens of embryos and young chicks express only the α-chain of CD8. Among intestinal epithelial lymphocytes, the major CD8+ T cell populations present in mice are conserved but there is a population of TCRγδ CD8αβ cells in the chicken that is not found in rodents. Chicken CD8 is expressed on approximately 80% of thymocytes, 15% of blood mononuclear cells, and 50% of spleen cells but less than 1% of cells in the bursa and bone marrow. The monoclonal antibody CT-8 recognizes the CD8α chain and has been shown to react to a polymorphic determinant in turkey.
