Description: CD90 (Thy-1), a GPI-anchored molecule and one of the smallest members of the immunoglobulin superfamily of cell surface receptors, consists of a single V-set domain. It is expressed on thymocytes, peripheral T lymphocytes, some intraepithelial T lymphocytes, and neurons of all mouse strains. The monoclonal antibody G7 stimulates T-cell proliferation and IL-2 secretion via signaling through the T-cell receptor/CD3 complex. G7 has also been reported to promote apoptosis of thymocytes and CTL clones and to mediate adhesion of thymocytes to thymic stroma.

Description: The CD45 antigen is an essential regulator of leukocyte activation and development and is now known to be a protein tyrosine phosphatase, e.g., CD45 is able to dephosporylate the Src family of protein kinases. This activity is known to be required for signal transduction induced by T and B cell antigen receptor engagement, cytokine signaling, and Fc receptor stimulation. The functional state of CD45 also has a pronounced effect on lymphocyte development. CD45 exists in multiple forms, such as the restricted variants (CD45R; see companion products 1660, 1665, 1675, 1685 and 1820) and as allelic forms CD45.1 (Ly-5.1) and CD45.2 (Ly-5.2). The specificity of antibodies to CD45.1 and CD45.2 has been exploited in studies where resolution of donor and recipient cells is essential, such as in gene therapy or bone marrow transplantation. Description: CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

Description: The lymphocyte Fcγ receptors recognize the Fc portion of IgG, presented either as immune complexes or as free antibody. The different classes of receptors are distinct because of varying size, tissue distribution and affinity for IgGs. The Fc type II receptor is expressed on a wide variety of cells including B cells, hematopoietic cells, monocyte/macrophages, neutrophils, platelets, Langerhans cells, eosinophils, basophils, trophoblasts, and endothelial cells of the placenta. The Fcγ type III receptors are higher affinity than the type II and are expressed on macrophages, NK cells and neutrophils. Both types of receptors can be expressed on the same cell and in varying ratios. The receptors are constitutively expressed, although cytokines and lymphokines can modulate their expression. Besides identifying FcγR+ cells, monoclonal antibodies to the FcγII/III receptor have been used to block Fc receptor binding of IgG, Fc-mediated signal transduction and effector functions, clearance of immune complexes and to attenuate infection by organisms dependent on FcγR for parasitic invasion. Description: The monoclonal antibody DX5 reacts with CD49b, also known as very late antigen-2 (VLA-2) and α2 integrin. The antibody stains the majority of NK cells and a small subpopulation of T cells in all mouse strains tested (e.g., A/J, AKR, BALB/c, C3H/HeJ, C57BL/6, C57BL/10, C57BR, C58, CBA/Ca, CBA/J, DBA/1, DBA/2, SJL, SWR). Three-color flow cytometry analysis has revealed that most cells that express NK-1.1 (NKR-P1C) also express CD49b. However, small subsets of DX5+NK-1.1- and DX5-NK-1.1+ cells can be found, especially among the CD3+ population of T lymphocytes. NK cells cultured in the presence of IL-2 progressively lose reactivity with DX5 as a consequence of cellular proliferation. Therefore, DX5 can be used to define functionally distinct subsets of murine NK cells. The monoclonal antibody DX5 has not been demonstrated to have activating or blocking activity.
