Description: CD4 is a type I transmembrane glycoprotein expressed on most thymocytes and the helper/inducer subpopulation of mature T lymphocytes of all mouse strains tested. CD4 has also been detected at low density on pluripotent hematopoietic stem cells, bone marrow myeloid precursors, and intrathymic lymphoid precursors. The CD4 antigen functions as a co-receptor, interacting with class II major histocompatability complex (MHC) molecules in the recognition of foreign antigens by T cells. In vivo and in vitro treatment with the GK1.5 monoclonal antibody effectively depletes CD4+ cells. GK1.5 also blocks helper T cell responses to MHC class II antigens, including cytolysis, proliferation, allogeneic B cell help, and release of lymphokines.

Description: Glial fibrillary acid protein (GFAP) is a member of the type III intermediate filament family of proteins. GFAP is heavily expressed in astrocytes and certain other astroglia in the central nervous system, in satellite cells in peripheral ganglia, and in non-myelinating Schwann cells in peripheral nerves. It is closely related to its non-epithelial family members, vimentin, desmin, and peripherin, which are all involved in the structure and functions of the cell’s cytoskeleton. GFAP is thought to help to maintain astrocyte mechanical strength, as well as the shape of cell; however, its precise function remains poorly understood. In adults, GFAP levels increase in response to the proliferation of astrocytes associated with Alzheimer’s disease, epilepsy, and multiple sclerosis. Antibodies specific for GFAP are useful as markers of astrocytic cells and neural stem cells as well as for distinguishing neoplasms of astrocytic origin from other neoplasms in the central nervous system.

Description: The lymphocyte Fcγ receptors recognize the Fc portion of IgG, presented either as immune complexes or as free antibody. The different classes of receptors are distinct because of varying size, tissue distribution and affinity for IgGs. The Fc type II receptor is expressed on a wide variety of cells including B cells, hematopoietic cells, monocyte/macrophages, neutrophils, platelets, Langerhans cells, eosinophils, basophils, trophoblasts, and endothelial cells of the placenta. The Fcγ type III receptors are higher affinity than the type II and are expressed on macrophages, NK cells and neutrophils. Both types of receptors can be expressed on the same cell and in varying ratios. The receptors are constitutively expressed, although cytokines and lymphokines can modulate their expression. Besides identifying FcγR+ cells, monoclonal antibodies to the FcγII/III receptor have been used to block Fc receptor binding of IgG, Fc-mediated signal transduction and effector functions, clearance of immune complexes and to attenuate infection by organisms dependent on FcγR for parasitic invasion. The monoclonal antibody 93 recognizes a conformational epitope formed by FcγII and FcγIII receptors.

Description: Ly-6A/E, also known as Sca-1, is a member of the Ly-6 multigene family of type V glycophosphatidylinositol-anchored cell surface proteins. It is expressed on multipotent hematopoietic stem cells in bone marrow of mice with both the Ly-6.1 and Ly-6.2 allotypes. In the periphery, Ly-6A/E exhibits a pattern of expression which is based on differences between the two allotypes. Ly-6.1 strains (e.g., A, BALB/c, CBA, C3H/He, DBA/1, NZB) have few Ly-6A/E+ resting peripheral lymphocytes whereas Ly-6.2 strains (e.g., AKR, C57BL, C57BR, C57L, DBA/2, PL, SJL, SWR, 129) have relatively high numbers of Ly-6A/E+ lymphocytes. The expression of Ly-6A/E is dramatically upregulated in all strains upon cellular activation. Studies with the monoclonal antibody D7 suggest that Ly-6A/E may be involved in B and T lymphocyte responses and it appears that the antigen may be required for T-cell receptor-mediated T-cell activation.

Along with CD80 (B7-1), CD86 participates in T cell costimulation via interaction with CD28 and CD152 (CTLA-4).
