Description: FAM19A4 is a secreted, 12 kDa member of the FAM19/TAFA family of chemokine-like proteins. Like other members of the FAM19/TAFA family, with the exception of TAFA5, mature FAM19A4 contains 10 regularly spaced cysteine residues. The FAM19A4 proteins are predominantly expressed in specific regions of the brain and the biological functions of FAM19A4 family members remain to be determined, but there are a few tentative hypotheses. First, FAM19A4 may modulate immune responses in the CNS by functioning as brain specific chemokines, and may act with other chemokines to optimize the recruitment and activity of immune cells in the CNS. Second, FAM19A4 may represent a novel class of neurokines that act as regulators of immune nervous cells. And third, FAM19A4 may control axonal sprouting following brain injury.

Description: Nucelobindin-2 (NUCB2; Nesfatin-1) was first isolated in adipocytes, but is also expressed by gastric mucosa and pancreatic beta cells. This pattern of nucleobindin-2 expression, as well as the presence of nucelobindin-2 within the plasma of rodents and humans, strongly suggests that this compound may act as a circulating regulatory factor. Nucelobindin-2 is reported to reduce food intake after administration. It is responsible for regulating appetite and production of body fat. Excess nucelobindin-2 in the brain leads to a loss of appetite, lack of nesfatin-1 in the brain leads to an increase of appetite.

Description: Nudel is important for normal cortical development. It is invovled in microtubule organization, nuclear translocation, and neuronal positioning in concert with various other factors (including Lis1, Pafah1b1, Pahfah1b2, dynein, dynorphin A and cdk5). Western blot analysis of mouse tissues shows abundant expression of Nudel in brain and testis, and much lower expression in heart, liver, kidney, and skeletal muscle. In fractionated rat brain, Nudel and Lis1 are both found in fractions enriched for postsynaptic density proteins. Immunostaining of embryonic day 18 mouse brain sections revealed staining of migrating neurons and thalamocortical axons of the intermediate zone of the developing cerebral cortex, as well as several other developing brain regions. The deduced protein contains 345 amino acids and has a calculated molecular mass of 38.4 kDa. It has a coiled coil motif (residues 19 to 201), followed by several potential phosphorylation sites for casein kinase II, protein kinase C or CDK5. Nudel shares about 50% identity with mouse and human NUDE proteins.
