Description: This MAb recognizes an extracellular epitope of an integral membrane glycoprotein of 150 kDa, identified as CD13. This antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. It is absent from normal lymphocytes, platelets and erythrocytes. CD13 is also present on fibroblasts; endothelial cells, epithelial cells from renal proximal tubules and intestinal brush border, bone marrow stromal cells, osteoclasts, and cells lining bile duct canaliculi. CD13 is identical to aminopeptidase N (APN), a prominent membrane-bound metalloprotease present on the surface of intestinal brush border and renal tubules. CD13 plays a role in metabolism of biologically active peptides, in phagocytosis, and in bactericidal/tumoricidal activities. It also serves as a receptor for human coronaviruses (HCV). The lineage-restricted pattern of expression of CD13 within the hemopoietic compartment suggests that it may be important in myeloid cell differentiation.

Description: This antibody reacts with five invariable CD3 chains designated CD3y, CD3d, CD3e, CD3f, and CD3n with molecular weight ranging from 16-28 kDa. It is found on all mature T-lymphocytes, NK, T-cells, and some thymocytes. CD3, also known as T3, is a member of the immunoglobulin superfamily that plays a role in antigen recognition, signal transduction, and T cell activation. It is expressed, typically at high levels, on peripheral T cells and majority of T cell neoplasms. Thymocytes express CD3 at different level on the cell surface in the course of differentiation and, in cortical thymus, CD3 is predominantly intracytoplasmic. This MAb is particularly useful for induction of T cell activation in vitro and in vivo due to its unusual IgG3 isotype.

Description: This antibody recognizes a protein of 33-55 kDa, identified as CD53 (Workshop VI; Code N-L033). It is expressed on monocytes and macrophages, dendritic cells, osteoblasts and osteoclasts, and on T and B cells from every stage of differentiation but is absent from platelets, red blood cells. CD53 appears to be the marker with widest reactivity as well as the marker with the strictest specificity to hematopoietic cells. CD53 is a type III membrane with both termini in the cytoplasm and two loops in the extracellular environment. This molecule, in common with other members of tetraspan family, is involved in cellular activation as part of a signal transduction complex involving other membrane glycoproteins. CD53 crosslinking induces calcium flux on human monocyte and B cells. Cross-linking of CD53 promotes activation of resting human B-lymphocytes. Description: This antibody recognizes a single chain glycoprotein of 105/120 kDa, identified as CD30/Ki-1. CD30 is synthesized as a 90 kDa precursor, which is processed in the Golgi complex into a membrane-bound phosphorylated mature 105/120 kDa glycoprotein. In Hodgkins disease, CD30/Ki-1 antigen is expressed by mononuclear-Hodgkin and multinucleated Reed-Sternberg cells. Description: This antibody recognizes a protein of 130-140 kDa, identified as CD22 (also known as BL-CAM). CD22 expression is restricted to normal and neoplastic B cells and is absent from other haemopoietic cell types. In B-cell ontogeny, CD22 is first expressed in the cytoplasm of pro-B and pre-B cells, and on the surface as B cells mature to become IgD . It is not expressed by plasma cells, CD22 is found highly expressed in follicular mantle and marginal zone B-cells, and while germinal center B-cells are relatively weak. Description: This antibody recognizes an intermediate filament protein (IFP) of 55 kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This MAb is highly useful in distinguishing ovarian carcinomas (keratin 7 ) from colon carcinomas (keratin 7-).
