Description: SIGIRR is a member of the Toll-like receptor-interleukin 1 receptor superfamily. Members of this family are defined by the presence of an intracellular Toll-IL-1R (TIR) domain. The Toll-like receptors (TLRs) are signaling molecules that recognize different microbial products during infection and serve as an important link between the innate and adaptive immune responses. SIGIRR was originally identified through database analysis and was shown to have only one Ig domain as opposed to the normal three Ig folds seen in the TIR family. Similar to ST2, another TIR family member, it has been shown to negatively regulate IL-1 receptor and Toll-like receptor signaling. However, SIGIRR inhibits TLR-IL-1R signaling by dimerizing with TLR4, TLR5, TLR9, and IL-1R. It also associates with the down-stream TLR signaling proteins IRAK and TRAF6 in an IL-1-dependent fashion.

Description: TCTN3 is a member of the Tectonic protein family, a group of evolutionarily conserved secreted and transmembrane proteins that regulate the Hedgehog (Hh)-mediated patterning of the neural tube. While the expression pattern and function of TCTN3 is not known, it is 58% similar to TCTN1. TCTN1 is expressed during embryonic development in regions that participate in Hh signaling, beginning in the gastrulation stages in the ventral node. Mice expressing mutant TCTN1 die between E13.5 and E16.5 and display holoprosencephaly, a defect associated with reduced Hh signaling, indicating the role of TCTN1 as an Hh activator. At later stages in development, TCTN1 is thought to also act as a repressor on the Hh pathway in the anterior and posterior neural tube.

Description: CD68, cluster of differentiation, is a 110-kD transmembrane glycoprotein that is highly expressed by human monocytes and tissue macrophages. CD68 is a member of a family of hematopoietic mucin-like molecules that includes leukosialin/CD43 and stem cell antigen CD34. The CD68 gene is mapped to 17p13.1. Immunohistochemistry can be used to identify the presence of CD68, which is found in the cytoplasmic granules of a range of different blood cells. It is particularly useful as a marker for the various cells of the macrophage lineage, including monocytes, histiocytes, giant cells, Kupffer cells, and osteoclasts. This allows it to be used to distinguish diseases of otherwise similar appearance, such as the monocyte/macrophage and lymphoid forms of leukaemia (the latter being CD68 negative). Its presence in macrophages also makes it useful in diagnosing conditions related to proliferation or abnormality of these cells, such as malignant histiocytosis, histiocytic lymphoma, and Gaucher's disease.
