Description: Influenza A virus is a major public health threat, killing more than 30,000 people per year in the USA (1). Novel influenza virus strains caused by genetic drift and viral recombination emerge periodically to which humans have little or no immunity, resulting in devastating pandemics. Influenza A can exist in a variety of animals, but it is in birds that all subtypes can be found (2). These subtypes are classified based on the combination of the virus coat glycoproteins hemagglutinin (HA) and neuraminidase (NA) subtypes. HA interacts with host cell surface proteins containing oligosaccharides with terminal sialyl residues. Its extracellular region has two domains (HA1 and HA2); HA1 is cleaved from the main hemagglutinin protein by the host immune system. During 1997, an H5N1 avian influenza virus was determined to be the cause of death in 6 of 18 infected patients in Hong Kong (3). This more recent virulent strain of H5N1 is now seen in Africa and Europe, as well as in Southeast Asia. There is some evidence of human to human spread of this virus, but it is thought that the efficiency of this type of transmission is low (4). Virus isolated from a human infected with the H5N1 strain in 1997 could bind to oligosaccharides from human as well as avian sources, indicating its species-jumping ability (5). This HA1 recombinant protein is recognized by several ProSci hemagglutinin antibodies.

Description: CD4 is a member of the immunoglobulin superfamily and is implicated as associative recognition element in MHC (Major Histocompatibility Complex) class II-restricted immune response (1). On T-lymphocytes, it defines the helper/inducer subset. The mature 55 kd CD4 protein consists of a 372 amino acid extracellular segment composed of four tandem immunoglobulin-like VJ regions. The CD4 molecule is a major receptor for human immunodeficiency virus (HIV), binding directly to the envelope glycoprotein gp120 on HIV, with the co-receptors being CCR5 or CXCR4. It has been shown that the V-like domains are critical for binding with HIV envelope gp120 (2). This soluble CD4 fragment (sCD4) will work in HIV neutralization assays at levels comparable to previously published results. The minor upper band seen in the Coomassie blue-stained gel is likely to be due to the presence of uncleaved signal sequence.

Description: Protoplast secreted protein 1 (Pst1) is an anchored plasma membrane protein. Pst1 was previously identified as a protein secreted by yeast regenerating protoplasts, which suggests a role in cell wall construction (1). Pst1 is a protein with 444 amino acids and is attached to yeast cell wall via a glycosylphosphatidylinisotol (GPI) anchor (2). Pst1 contains 15 potential N-linked glycosylation sites and is heavily glycosylated. It migrates at around 200 kDa on SDS-PAGE when produced in wild type S. cerevisiae. Our Pst1 protein was expressed in a genetically manipulated triple-mutant (TM) S. cerevisiae stain (Δoch1 Δmnn1 Δmnn4), which results in the production of sole Man8GlcNAc2 carbohydrate structures and Pst1 migration at approximately 100 kDa. When produced in the TM yeast, Pst1 can be recognized by several HIV-1 broadly neutralizing antibodies, including 2G12 and recently identified glycan-specific PGT antibodies (3). Among several heavily N-glycosylated TM yeast glycoproteins, Pst1 shows high affinity for 2G12 and efficiently inhibits gp120 interactions with 2G12 and DC-SIGN, and it also blocks 2G12-mediated neutralization of HIV-1 pseudoviruses (4).
