Description: TBC1D1 is the founding member of a family of proteins sharing a 180- to 200-amino acid TBC domain and presumed to have a role in regulating cell growth and differentiation. These proteins share significant homology with TRE2/USP6, yeast Bub2, and CDC16. TBC1D1 and TBC1D4 (AS160) have been demonstrated to be Rab GAPs (GTPase-activating proteins) that link upstream to Akt and phosphoinositide 3-kinase and downstream to Rabs involved in trafficking of GLUT4 vesicles. TBC1D1 regulates insulin-mediated GLUT4 translocation through its GAP activity, and is a likely Akt substrate. Mutations in the Tbc1d1 gene lead to some cases of severe human obesity.

Description: BICD2 is the second human homolog discovered to the Drosophila Bicaudal-D protein that forms part of the cytoskeleton and mediates the correct sorting of mRNAs for oocyte- and axis-determining factors during oogenesis. Description: Thymic stromal lymphopoietin (TSLP) has recently been identified as an important factor capable of driving dendritic cell maturation and activation. TSLP is a four-helix-bundle cytokine that is expressed mainly by barrier epithelial cells and is a potent activator of several cell types such as myeloid dendritic cells. TSLP is involved in the positive selection of regulatory T cells, maintenance of peripheral CD4+ T cell homeostasis and the induction of CD4+ T cell-mediated allergic reaction. TSLP is also capable of supporting the growth of fetal liver and adult B cell progenitors and their differentiation to the IgM-positive stage of B cell development. Amino acid sequence analysis has shown poor homology between human and mouse TSLP although they exhibit similar biological functions and are expressed in similar tissues. At least two differentially spliced isoforms of TSLP are known to exist.
