Description: TANK was initially identified as a novel TRAF-interacting protein that regulated TRAF-mediated signal transduction. Specifically, ligand binding by surface receptors in the tumor necrosis factor (TNF) receptor and Toll/interleukin-1 (IL-1) receptor families lead to the formation of a TRAF/TANK complex that mediates the activation of the transcription factor NF-kB. This activation of NF-kB occurs through an association with the kinases IKKe and TBK1. More recently, it was shown that these proteins can then form a complex with NEMO, a protein that regulates the activity of the IkB complex. This suggests that in addition to the possibility that TBK1 and IKKe activate the IKKs, the association with the IKK complex may help these kinases modulate other functions, such as the transactivation potential of NF-kB proteins. At least two isoforms of TANK are known to exist.

Description: TNF-related weak inducer of apoptosis (TWEAK) is a member of the tumor necrosis factor superfamily (TNFSF) of structurally related cytokines. Like most other members of this family, TWEAK is a cell surface-associated type II transmembrane protein although a smaller, biologically active form can also be generated by cleavage near the cell membrane. TWEAK has multiple biological activities, including stimulation of cell growth and angiogenesis, induction of inflammatory cytokines, in addition to stimulation of apoptosis. The TWEAK signal transduction pathway has not been well established but it appears to signal via TweakR/Fn14 in a manner similar to that described for other TNFSF members that bind receptors lacking death domains.

Description: Phosphoribosyl transferase domain containing 1 (PRTFDC1) is highly homologous to the hypoxanthine phosphoribosyltransferase (HPRT1) and may have arisen from a gene duplication event of a common ancestor gene. Recently, it was shown that CpG islands in the PRTFDC1 promoter could be hypermethylated in ovarian cancers and oral squamous-cell carcinomas (OSCC), leading to gene silencing. Description: SCUBE3 is a member of a family of secreted glycoproteins that contain N-terminal EGF-like repeats and C-terminal cysteine-rich motifs and CUB domain and is highly expressed in primary osteoblasts and bones, and to a lesser extent in heart (1,2). Other studies have shown that overexpression of SCUBE3 in mice induced cardiac hypertrophy, suggesting that it may also play a role in the regulation of cardiac growth. SCUBE3 has been shown to be an endogenous TGF-beta receptor ligand (3,4) and is thought to promote lung cancer cell mobility and invasiveness. In lung cancer cells, the secreted SCUBE3 protein was cleaved by MMP2 and MMP9, allowing the activation of the TGF-beta receptor, the increase of Smad2/3 transcriptional activity and the upregulation of expression of proteins such as TGF-beta1, VEGF, Snail, and Slug.
