Description: The Lewis histo-blood group system comprises a set of fucosylated glycosphingolipids that are synthesized by exocrine epithelial cells and circulate in body fluids. The glycosphingolipids function in embryogenesis, tissue differentiation, tumor metastasis, inflammation, and bacterial adhesion. They are secondarily absorbed to red blood cells giving rise to their Lewis phenotype. This gene is a member of the fucosyltransferase family, which catalyzes the addition of fucose to precursor polysaccharides in the last step of Lewis antigen biosynthesis. It encodes an enzyme with alpha(1,3)-fucosyltransferase and alpha(1,4)-fucosyltransferase activities. Lewis blood group antigens are carbohydrate moieties structurally integrated in mucous secretions. Lewis antigen system alterations have been described in gastric carcinoma and associated lesions. Anomalous expression of Lewis B antigen has been found in some non-secretory gastric carcinomas and colorectal cancers.

Description: This antibody recognizes a protein of 32 kDa, identified as CD8a (also known as CD8 chain, T cell co-receptor, Leu2, and T8). CD8 molecule consists of two chains, termed and chain, which are expressed as a disulphide-linked heterodimer or as an homodimer. CD8 is expressed on T cell subset (cytotoxic/suppressor T cells), thymocytes and NK cells. The majority of CD8 T-cells expresses CD8 as heterodimer. Some subpopulation of CD8 T cells as well as NK cells may express homodimer. CD8 functions as a co-receptor in concert with TCR for binding the MHC class I/peptide complex. The HIV-2 envelope glycoprotein binds CD8 chain (but not chain). The cytoplasmic domain of CD8 associates with p56lck tyrosine kinase.

Description: This antibody recognizes a protein of 30-33 kDa, which is identified as CD20 (Workshop V; Code CD20.4). It is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. The protein passes through the membrane 4 times with both ends in cytoplasm and exposes one short and one longer loop to the external environment. CD20 is not glycosylated in resting B-cells and its cytoplasmic domains are differentially phosphorylated upon activation. It acts as calcium channel involved in B cell activation and cell cycle progression.

Description: This MAb reacts with MUC1. The dominant epitope of this MAb has not yet been determined. MUC1 is a large cell surface mucin glycoprotein expressed by most glandular and ductal epithelial cells and some hematopoietic cell lineages. It is expressed on most secretory epithelium, including mammary gland and some hematopoietic cells. It is expressed abundantly in lactating mammary glands and over expressed abundantly in >90% breast carcinomas and metastases. Transgenic MUC1 has been shown to associate with all four c-erbB receptors and localize with c-erbB1 (EGFR) in lactating glands. The MUC1 gene contains seven exons and produces several different alternatively spliced variants. The major expressed form of MUC1 uses all seven exons and is a type 1 transmembrane protein with a large extracellular tandem repeat domain. The tandem repeat domain is highly O glycosylated and alterations in glycosylation have been shown in epithelial cancer cells. Antibody to EMA is useful as a pan-epithelial marker for detecting early metastatic loci of carcinoma in bone marrow or liver.

Description: This antibody recognizes a single protein of 33-34 kDa, identified as the prostate specific antigen (PSA). This MAb is highly specific to PSA and stains prostatic secretory and ductal epithelium in both normal and neoplastic tissues. PSA is a chymotrypsin-like serine protease (kallikrein family) exclusively produced by the prostate epithelium, and abundant in seminal fluid. PSA can be detected in the sera of patients with prostatic carcinoma. It is predominantly complexed to a liver-derived serine protease inhibitor, alpha-1-antichymotrypsin (ACT). A higher proportion of serum PSA is complexed to ACT in prostate cancer than in benign prostate hyperplasia.

Description: This antibody recognizes a protein of 36 kDa, identified as Thymidylate Synthase (TS) (EC 2.1.1.45). TS converts deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), which is essential for DNA biosynthesis. TS is also a critical target for the fluoropyrimidines, an important group of antineoplastic drugs that are widely used in the treatment of solid tumors. Both 5-FU and fluorodeoxyuridine are converted in tumor cells to FdUMP which inactivates TS by formation of a ternary covalent complex in the presence of the folate cofactor 5,10-methylenetetrahydrofolate. Expression of TS protein is associated with response to 5-fluorouracil (5-FU) in human colorectal, gastric, head and neck, and breast carcinomas.
