Description: The monoclonal antibody 2F1 reacts with mouse killer cell lectin-like receptor G1 (KLRG1; formerly known as mouse MAFA), a homodimeric member of the lectin-like type 2 transmembrane receptor family that contain characteristic immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in their cytoplasmic domains. These ITIMs interact with the SH2 domains of protein phosphatases such as SHP-1. The 2F1 antibody stains 30-60% of NK1.1+CD3- splenocytes, and a small fraction of T cells in all mouse strains tested (C57BL/6, BALB/c, 129/J, C3H.SW, AKR/J, SJL). Cell surface expression of KLRG1 is up-regulated by expression of MHC class I molecules. The effect of MHC class I expression is indirect, and can be mediated by interactions with class I-specific Ly49 inhibitory receptors.

Description: Keratins are a family of intermediate filament proteins that assemble into filaments through forming heterodimers of one type I keratin (keratins 9 to 23) and one type II keratin (keratins 1-8). The two keratin types share only 30% sequence homology. Keratins demonstrate tissue- and differentiation-specific expression profiles. Cytokeratin 19 is the smallest human keratin. It is found in many simple and some non-keratinizing stratified squamous epithelia and is often co-expressed with keratin 7. Cytokeratin 19 has been implicated as a marker for stem cells in some tissues such as hair follicles in the skin. Monoclonal antibodies to cytokeratin proteins can be useful markers for tumor identification and classification. The degree of cytokeratin 19 positivity in breast cancer distinguishes malignant from benign tumors. Cytokeratin 19 is also associated with malignant transformation in esophageal and pancreatic cancers.

Description: The CD117 (c-Kit) antigen is the cell-surface receptor for stem cell factor (SCF). Together these molecules constitute a ligand/receptor pair which functions to maintain normal hematopoiesis in the adult. Signaling through SCF/c-Kit has an important role in stimulating myeloid and erythroid production of primitive hematopoietic progenitor cells. A variety of cytokines act synergistically with SCF to stimulate proliferation and differentiation of bone marrow progenitor cells. For example, SCF plus IL-7 can stimulate the combined myeloid and B cell differentiation of uncommitted hematopoietic progenitor cells. SCF/c-Kit also participates in erythropoiesis in both the bone marrow and spleen. Loss-of-function mutations for SCF/c-Kit lead to a variety of pleiotropic developmental defects while gain-of-function mutations can lead to constitutive activation of the kit receptor and links to cancer. Examples of the former are mast cell deficiency and severe macrocytic anemia while the latter mutation has been identified in mastocytomas.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow.
