Description: This MAb recognizes a protein of 145 kDa, identified as CD117/p145kit. It is found on a wide variety of tumor cells including follicular and papillary carcinoma of thyroid, adenocarcinomas from endometrium, lung, ovary, pancreas, and breast as well as malignant melanoma, endodermal sinus tumor, and small cell carcinoma. However, anti-CD117 has been particularly useful in differentiating gastrointestinal stromal tumors from Kaposi's sarcoma, tumors of smooth muscle origin, fibromatosis, and neural tumors of the GI tract. Anti-CD117 is also useful in recognizing myeloblasts in bone marrow biopsy and clot section.

Description: This antibody recognizes a protein of 36 kDa, identified as cyclin D1. Cyclin D1, one of the key cell cycle regulators, is a putative proto-oncogene overexpressed in a wide variety of human neoplasms. This antibody neutralizes the activity of cyclin D1 in vivo. About 60% of mantle cell lymphomas (MCL) contain a t(11; 14)(q13; q32) translocation resulting in over-expression of cyclin D1. This antibody is useful in identifying mantle cell lymphomas (cyclin D1 positive) from CLL/SLL and follicular lymphomas (cyclin D1 negative). Occasionally, hairy cell leukemia and plasma cell myeloma weakly express Cyclin D1.

Description: This antibody recognizes a 66 kDa protein, which is identified as Tripartite motif-containing protein 29 (TRIM29). It interacts with the intermediate filament protein vimentin, a substrate for the PKC family of protein kinases, and with hPKCI-1, an inhibitor of the PKCs. TRIM29 protein contains both zinc finger and leucine zipper motifs, suggesting that the it may form homodimers and possibly associate with DNA. High expression of TRIM29 has been reported in gastric cancer and pancreatic cancer, and correlates with enhanced tumor growth and lymph node metastasis. Description: This antibody recognizes a protein of ~55 kDa, identified as SOX10. This MAb is highly specific and does not cross-react with other members of the SOX-family. SOX genes comprise a family of genes that are related to the mammalian sex-determining gene SRY. These genes similarly contain sequences that encode for the HMG-box domain, which is responsible for the sequence-specific DNA-binding activity. SOX-10 is a sensitive marker of melanoma, including conventional, spindled, and desmoplastic subtypes. It is expressed by metastatic melanomas and nodal capsular nevus in sentinel lymph nodes, but not by other lymph node components such as dendritic cells, which usually express S100 protein. Commonly used melanoma markers, such as anti-HMB-45 and anti-Melan-A, are poorly expressed in desmoplastic melanomas while SOX-10 is moderately to strongly expressed in desmoplastic melanomas. SOX-10 is considered as a very reliable marker for recognizing residual desmoplastic melanomas. In normal tissues, it is expressed in Schwann cells, melanocytes, and myoepithelial cells of salivary, bronchial and mammary glands. SOX-10 expression is also observed in mast cells.
