Description: CD62L, also known as L-selectin, is a type I transmembrane glycoprotein with lectin-like and epidermal growth factor-like domains. It binds to sialylated oligosaccharide determinants on high endothelial venules (HEV) in peripheral lymph nodes. In the mouse, CD62L is expressed on most thymocytes and on peripheral leukocytes, including B and T lymphocytes, neutrophils, monocytes, and eosinophils. This member of the selectin family of cell adhesion molecules appears to be required for lymphocyte homing to peripheral lymph nodes and for leukocyte extravasation at sites of inflammation. Expression of CD62L on lymphocytes and neutrophils is rapidly down-regulated upon cell activation and the level of CD62L expression, along with other markers, distinguishes naive T cells from effector/memory T cells. The monoclonal antibody MEL-14 blocks in vitro binding of lymphocytes to peripheral lymph node HEV and inhibits in vivo lymphocyte extravasation into peripheral lymph nodes and the late stages of leukocyte rolling.

Description: CD80 (B7-1) is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed by dendritic cells, monocytes, and peritoneal macrophages. Expression of CD80 on B cells is upregulated after activation by various means including stimulation by LPS, IL-4, and surface Ig receptor crosslinking with peak expression occurring between 48 and 72 hours post-activation. The interaction of CD80 with its ligands, CD28 and CD152 (CTLA-4), provides a potent costimulatory signal for T cells activated through the CD3/T cell receptor complex. The monoclonal antibody 1G10 blocks binding of CTLA-4 to CD80 but it does not block stimulation of T cells by natural antigen-presenting cells.

Description: CD19 is a monomeric transmembrane glycoprotein expressed at relatively constant levels throughout B cell development from early pro-B/pre-B cells (i.e. B220+/CD43+/HSA+) through fully differentiated B cell stages. Terminally differentiated plasma cells do not express CD19. In humans, the CD19 molecule on the surface of mature B cells associates with CD21 (CR-2) and CD81 (TAPA-1), and this multimolecular complex synergizes with surface immunoglobulin to provide signal transduction and promote cellular activation. All splenic and peritoneal IgM+ cells of both B-1 and B-2 lineages are CD19+, with B-1 cells expressing higher levels of CD19 than B-2 cells in these sites. Studies with CD19-deficient mice have suggested that this molecule may not be required for normal generation and maturation of B cells in the bone marrow. The monoclonal antibody 6D5 recognizes the same epitope as the published clone, 1D3.

Description: CD49d, a member of the integrin family of extracellular matrix and cell adhesion molecules, is the integrin α4 chain. CD49d associates noncovalently with CD29, the integrin β1 subunit, to form the α4β1 very late antigen-4 (VLA-4) heterodimer. Integrin α4 can also associate with integrin β7 to form the α4β7 heterodimer. It is expressed on most leukocytes with the possible exception of neutrophils. VLA-4 mediates leukocyte binding to CD106 (VCAM-1) and is involved in the migration of leukocytes from blood to tissue at sites of inflammation. VLA-4 can also provide a costimulatory signal to T cells for activation and proliferation. The α4β7 integrin heterodimer is expressed on a small subpopulation of thymocytes, most lymph node T and B lymphocytes, NK cells, and eosinophils. Integrin α4β7 also binds to fibronectin and VCAM-1.

Description: CD95, also known as Fas and APO-1, is a 45 kDa type I transmembrane glycoprotein and a member of the tumor necrosis factor receptor superfamily. It is expressed by activated lymphocytes, monocytes, neutrophils, fibroblasts, and cell lines. Fas ligand binding to CD95 induces apoptosis in activated mature lymphocytes thereby playing a role in maintaining peripheral tolerance. Crosslinking of CD95 by the monoclonal antibodies DX2 and DX3 delivers an apoptotic signal to Fas-sensitive cells indicating that these monoclonal antibodies recognize a functional epitope of CD95.
