Description: CD107b, also known as lysosomal-associated membrane protein 2 (LAMP-2), is a heavily glycosylated type I transmembrane protein that constitutes the major sialoglycoproteins on lysosomal membranes. It is a ligand for galaptin, an S-type lectin present in extracellular matrix, through its recognition of acetyllactosamine oligosaccharide chains and is a ligand for E-selectin-mediated cell adhesion. CD107b is expressed in lysosomal/endosomal membranes and on the surface of activated platelets and some tumor cell lines including U937 and KG1a. LAMP-2 may function in protecting the inner surface of the lysosomal membrane by forming a barrier to lysosomal hydrolases. The upregulation of both CD107a and CD107b on the surface of tumor cell lines has been associated with their enhanced metastatic potential where they may increase adhesion to extracellular matrix and endothelium.

Description: The monoclonal antibody TCR-2 precipitates a CD3-associated heterodimer of Mr 90 kDa (two bands of Mr 50 kDa and 40 kDa upon reduction) on chicken peripheral blood T cells. Deglycosylation of the heterodimer yields two polypeptides of Mr 34 kDa and 29 kDa. In the chicken, two distinct subpopulations of αβ T cells appear in the thymus subsequent to the appearance of γδ T cells. These subpopulations, originally denoted as TCR2 and TCR3, arise sequentially in the thymus during ontogeny and are now known to represent two distinct Vβ families, Vβ1 and Vβ2, respectively. The TCR-2 monoclonal antibody reacts with approximately 40% of thymocytes, 40-50% of blood mononuclear cells, and 40% of splenocytes in the chicken. Two-color immunofluorescence has revealed that most of the TCR2+ thymocytes express both CD4 and CD8 antigens. The TCR2+ cells in blood were found to express either CD4 (74 ± 2%) or CD8 (26 ± 4%). TCR2+ cells in the spleen also express either CD4 (37 ± 1%) or CD8 (64 ± 4%). Surprisingly, a relatively large subpopulation of CD8+ cells in the spleen are negative for TCR2. This observation led to the demonstration that 71 ± 6% of the TCRγδ+ cells in the spleen express CD8.

Description: CD80 (B7-1) is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is constitutively expressed by dendritic cells, monocytes, and peritoneal macrophages. Expression of CD80 on B cells is upregulated after activation by various means including stimulation by LPS, IL-4, and surface Ig receptor crosslinking with peak expression occurring between 48 and 72 hours post-activation. The interaction of CD80 with its ligands, CD28 and CD152 (CTLA-4), provides a potent costimulatory signal for T cells activated through the CD3/T cell receptor complex. The monoclonal antibody 1G10 blocks binding of CTLA-4 to CD80 but it does not block stimulation of T cells by natural antigen-presenting cells.

Description: The monoclonal antibody 34-5-8S reacts with a conformational epitope on H-2Dd MHC Class I found on the N-terminal domains of α1 and α2 chains when complexed with β2-microglobulin. The antibody does not react with H-2Dd α chains synthesized in vitro. Weak cross-reactivity with cells from mice of the H-2b, H-2q, and H-2s haplotypes has been observed by flow cytometric analysis. Reactivity with cells from mice of the H-2f, H-2k, H-2p, and H-2r haplotypes has not been observed. 34-5-8S has been reported to block the recognition of H-2Dd by Ly-49A+, Ly-49C+, and Ly-49G2+ natural killer cells.
