Description: Steroid receptor co-activators (SRCs) were initially described as nuclear receptor transcription co-activators, but they have recently been determined to co-regulate transcription initiated by other transcription factors. GAS is a recently identified glutamate-rich protein that interacts with SRC1, but not GRIP1 or AIB1, the other two members of the SRC family. GAS can also interact with the alpha subunit of the estrogen receptor (ERalpha), but not other receptors such as the retinoic acid receptor a, suggesting the interaction between GAS and ERalpha is relatively specific. Depletion of GAS by RNA interference in MCF7 cells led to a decrease in the mRNA and protein levels of ER target genes such as pS2, c-Myc and cyclin D1, indicating the role of GAS in the regulation of ER target genes. GAS has also been found to associate with an SET1-like methyltransferase complex specific for H3K4 methylation, suggesting that GAS has multiple roles in transcriptional regulation.

Description: Apoptosis is related to many diseases and development. Members in the Bcl-2 family are critical regulators of apoptosis by either inhibiting or promoting cell death. Bcl-2 homology 3 (BH3) domain is a potent death domain. BH3-only proteins, including Bad, Bid, Bik, Hrk, Bim, Noxa, and PUMA, form a growing subclass of the Bcl-2 family. A novel BH3-only protein was recently identified in human and mouse and designated Bmf (for Bcl-2-modifing factor). The BH3 domain in Bmf is required both for binding to Bcl-2 proteins and for triggering apoptosis. In healthy cells, Bmf associates with the dynein light chain 2 (DLC2) component of the myosin V motors and is sequestered by the cell's actin cytoskeleton. Disruption of the actin cytoskeleton, either by depolymerization of actin filaments or by detachment of cells from the extracellular matrix, triggers release and activation of Bmf, initiating the downstream apoptotic program. Bmf is constitutively expressed in many tissues.

Description: α-Actinin-4 is an actin-bundling protein of ~100kDa that is associated with cell motility, endocytosis, and cancer invasion. The α-actinin family comprises two non-muscle isoforms (α-actinin-1 and -4) and two skeletal muscle isoforms (α-actinin-2 and -3), with α-actinin-2 being also expressed in cardiac muscle. While α-actinin-4 is almost ubiquitously expressed, particularly high concentrations are found in glomeruli. On the subcellular level it is associated with actin stress fibers, but in certain cells it also localizes to the nucleus. Mutations in the α-actinin-4 gene cause an autosomal-dominant form of familial focal segmental glomerulosclerosis (FSGS), which is thought to result from a defect in glomerular podocyte function. A point mutation in the α-actinin-4 gene was found to generate an antigenic peptide that is recognized by autologous cytolytic T lymphocytes (CTL) on a human lung carcinoma. α-actinin-4 interacts with a variety of proteins, including the ring finger protein BERP, the PDZ-LIM protein CLP-36, the hemidesmosomal and cell-cell contact protein BP180, and the tight junction protein MAGI-1.
