Description: ImmunoReagents’ Biotinylated antibody conjugates are widely used due to their ability to enhance antibody signal in most immunoassays. Biotinylated secondary antibodies are used in most immunoassay formats (i.e., ELISA, Immunohistochemistry (IHC), and Western blotting). While Biotinylated antibodies by themselves do not produce a signal that can be visualized, biotin has a high affinity and specificity to Streptavidin/Avidin. Using ImmunoReagents’ streptavidin conjugates with biotinylated antibodies results in signal amplification.

Description: CAPS2 and the related protein CAPS1 encode novel neural/endocrine-specific cytosolic and peripheral membrane proteins. Both are essential components of the synaptic vesicle priming machinery and are required for the Ca2+-regulated exocytosis of secretory vesicles; CAPS-deficient neurons contain no or very few fusion competent synaptic vesicles, causing a selective impairment of fast phasic transmitter release. Like CAPS1, numerous isoforms of CAPS2 are known to exist. These isoforms show distinct expression patterns in the brain. For example, CAPS2b showed high expression in the developing cerebellum. Furthermore, one version of CAPS2 mRNA that lacks exon 3 is detected in some autistic patients, suggesting that the differential expression pattern of CAPS2 is involved in neuronal development.

Description: Tumor necrosis factor receptor superfamily member 1A (TNF p55 Receptor) is a receptor for TNFSF2/TNF-alpha and homotrimeric TNFSF1/lymphotoxin-alpha. The adapter molecule FADD recruits caspase-8 to the activated receptor. The resulting death-inducing signaling complex (DISC) performs caspase-8 proteolytic activation which initiates the subsequent cascade of caspases (aspartate-specific cysteine proteases) mediating apoptosis. It contributes to the induction of non-cytocidal TNF effects including anti-viral state and activation of the acid sphingomyelinase.

Description: IL-36A is a member of the interleukin 1 cytokine family whose gene and eight other interleukin 1 family genes form a cytokine gene cluster on chromosome 2 (1). IL-36A is thought to activate the NF-kappaB pathway through IL-1 receptor family members IL-1RL2 and IL-1RAcP (2). Like the related proteins IL-36B and IL-36G, IL-36A requires post-translational processing for full agonist activity, but the cleavage mechanism is currently unknown (3). The IL-36 cytokines have been suggested to amplify Th1 responses by enhancing proliferation and Th1 polarization of naive CD4+ T cells (4).

Description: SPATA18, also known as SPETEX-1, was initially identified as a cytoplasmic protein of elongate spermatids that is thought to be involved in differentiation process of elongate spermatids to mature spermatozoa. Recent experiments have shown that SPATA18 is a transcriptional target of p53 and p63. It has also been suggested to play a role in mitochondria quality control by inducing intramitochondrial lysosome-like organella that eliminate oxidized mitochondrial proteins.

Description: Vesicle-associated membrane protein (VAMP), also known as synaptobrevin, is an 18 kDa integral membrane protein localized to the cytoplasmic surface of synaptic vesicles. Two VAMP homologs, VAMP1 and VAMP2, are differentially expressed in the nervous system. VAMP2 is found in neurons associated with autonomic, sensory and integrative functions, and in non-neuronal Langerhans islets and glomerular cells. VAMP is a component of a 20S complex implicated in vesicle docking and fusion, the formation of which involves the interaction of VAMP with syntaxin through an ATP-dependent reaction modulated by synaptotagmin, SNAP-25, alpha-SNAP, and NSF.

Description: The MAK10 gene encodes a 733-amino acid protein with several regions of similarity to T cell receptor alpha-subunit V (variable) regions in yeast. The mammalian homologue of yeast MAK10, also known as EGAP, is one subunit of a novel N-terminal acetyltransferase (NAT) that is highly conserved among vertebrate species. It is expressed in a variety of tissues in the developing rat embryo but restricted in expression in the adult, remaining detectable only in tissues undergoing continual cell renewal or in cells responding to pathological injury. The MAK10-NAT complex is an essential regulatory enzyme controlling the function of a subset of proteins required for embryonic growth control and vessel development. This complex functionally co-assembles in mammalian cells to regulate cell proliferation and is essential for embryonic development, at least in part through the regulation of target of rapamycin (TOR) signaling events. At least two isoforms of MAK10 are known to exist.

Description: The OKT4 antibody reacts with human CD4, a 59 kDa protein which acts as a co-receptor for the T cell receptor (TCR) in its interaction with MHC Class II molecules on antigen-presenting cells. The extracellular domain of CD4 binds to the beta-2 domain of MHC Class II, while its cytoplasmic tail provides a binding site for the tyrosine kinase lck, facilitating the signaling cascade that initiates T cell activation. CD4, and co-receptors CCR5 and CXCR4, may also be utilized by HIV-1 to enter T cells. Human CD4 is typically expressed on thymocytes, some mature T cell populations such as Th17 and T regulatory (Treg) cells, as well as on dendritic cells.

Description: Alpha-crystallins, which are part of the small Heat shock family members, are major water-soluble proteins present in the lens of the mammalian eye. Phosphorylation of serine residues which occurs during development and in response to stress, is intimately linked with its function. Chaperone activity requires, and is modulated by, oligomerization and is limited to binding unfolded intermediates to prevent irreversible aggregation.

Description: PINK1 (PTEN induced putative kinase 1) is a mitochondrial serine/threonine kinase which maintains mitochondrial function/integrity, provides protection against mitochondrial dysfunction during cellular stress, potentially by phosphorylating mitochondrial proteins, and is involved in the clearance of damaged mitochondria via selective autophagy (mitophagy). PINK1 is synthesized as a 63 kD protein which undergoes proteolyt processing to generate at least two cleaved forms (55 kD and 42 kD). PINK1 and its substrates have been found in the cytosol as well as in different sub-mitochondrial compartments, and according to the recent reports; PINK1 may be targeted to OMM (outer mitochondrial membrane) with its kinase domain facing the cytosol, providing a possible explanation for the observed physical interaction with the cytosolic E3 ubiquitin ligase Parkin.
Defective PINK1 may cause alterations in processing, stability, localization and activity as well as binding to substrates/interaction-partners which ultimately leads to differential effects on mitochondrial function and morphology. Mutations in PINK1 are linked to autosomal recessive early onset Parkinson's disease, and are associated with loss of protective function, mitochondrial dysfunction, aggregation of alpha-synuclein, as well as proteasome dysfunction. Areas of interest and use for researchers include Neuroscience, mitochondrial function, and CDK-mediated phosphorylation pathways.

Description: The ACK2 antibody is specific for CD117, also called c-Kit, a 145 kDa cytokine receptor important in the development of hematopoietic stem cells, in oogenesis, and for functional activity of immune cells such as NK and mast cells. c-Kit binds to a ligand known as stem cell factor (SCF), or alternatively as mast cell growth factor. Ligand binding promotes the activation (dimerization) and subsequent tyrosine kinase activity of the c-Kit receptor and triggers key survival, expansion and maturation signals during hematopoietic progenitor cell development. Conversely, shedding of extracellular domain of c-Kit receptor is reported to induce inactivation or apoptosis within these cells. The survival signaling activity of c-Kit confers a proto-oncogenic attribute to the receptor, as overexpression or mutations in this protein are associated with tumor development.

Description: SLIT and NTRK-like family 1 (Slitrk1) is a member a protein family consisting of six homologous transmembrane proteins (Slitrk1-6) that share two conserved leucine-rich repeat domains in the extracellular domain and have significant homology to Slit, a secreted axonal growth-controlling protein. These proteins are also homologous to trk neurotrophin receptors in their intracellular domains. Expression of Slitrk proteins is highly restricted to neural and brain tumor tissues, but varies within the family. For example, Slitrk1 is expressed primarily in mature neurons. Overexpression of Slitrk1 in transfected neuronal cells induced unipolar neurites, while expression of the other Slitrk proteins inhibited neurite outgrowth, suggesting that these proteins are involved in the control of neurite outgrowth. While Slitrk1 variants have been suggested associated with Tourette's Syndrome, it is thought to play only a minor role if at all.
