Description: SIRT5 is the only human class III sirtuin and as such is the closest homolog to bacterial (e.g. E. coli CobB) and archaeal sirtuins. SIRT5 is an imported mitochondrial protein with a calculated precursor molecular weight of 34kDa. In various human, rat, mouse and bovine cells and tissues, the major reactive band migrates with an apparent molecular weight of ~29kDa. This presumably reflects removal of an N-terminal transit sequence due to mitochondrial import and processing. No cross-reactivity observed with recombinant human sirtuins SIRT1, SIRT2 or SIRT3.

Description: Autophagy, the process of bulk degradation of cellular proteins through an autophagosomic-lysosomal pathway is important for normal growth control and may be defective in tumor cells. It is involved in the preservation of cellular nutrients under starvation conditions as well as the normal turnover of cytosolic components and is negatively regulated by TOR (Target of rapamycin). PIST, a PDZ-containing protein, was discovered in a yeast two-hybrid system as a binding partner to Beclin-1, a Bcl-2-interacting protein homologous to the yeast autophagy gene apg6. Experiments with mutant PIST proteins lacking the PDZ domain showed that PIST interaction with Beclin-1 through its coiled-coil domain can modulate Beclin-1 activity and suggest that PIST interactions with other proteins through its PDZ domain may regulate the activity of PIST and Beclin-1.

Description: Aldh5A1 is a member of the aldehyde dehydrogenase superfamily, a group of NAD(P)(+)-dependent enzymes that catalyze the oxidation of a wide spectrum of aliphatic and aromatic aldehydes. Aldehyde dehydrogenase enzymes are thought to play a major role in the detoxification of aldehydes generated by alcohol metabolism and lipid peroxidation. Aldh5A1 is a mitochondrial NAD(+)-dependent succinic semialdehyde dehydrogenase. A deficiency of this enzyme, known as 4-hydroxybutyricaciduria, results in a disorder of the neurotransmitter 4-aminobutyric acid (GABA). Symptoms usually include static encephalopathy, associated with developmental delays, hypotonia, ataxia, speech defects, and seizures. At least two isoforms of Aldh5A1 are known to exist.

Description: Apoptosis is related to many diseases and development. The p53 tumor-suppressor protein induces apoptosis through transcriptional activation of several genes. A novel p53 inducible pro-apoptotic gene was identified recently and designated PUMA (for p53 upregulated modulator of apoptosis) and bbc3 (for Bcl-2 binding component 3) in human and mouse. PUMA/bbc3 is one of the pro-apoptotic Bcl-2 family members including Bax and Noxa, which are also transcriptional targets of p53. The PUMA gene encodes two BH3 domain-containing proteins termed PUMAa and PUMAb. PUMA proteins bind Bcl-2, localize to the mitochondria, and induce cytochrome c release and apoptosis in response to p53. PUMA may be a direct mediator of p53-induced apoptosis.
