Description: ZNF346, also known as JAZ (just another zinc-finger protein), is a nucleolar, zinc finger protein which preferentially binds to double-stranded (ds) RNA or RNA/DNA hybrids with high affinity via C2H2 zinc fingers. ZNF346 contains four CH-type zinc finger motifs that are connected by long (28-38) amino acid linker sequences. ZNF346 is expressed in all tissues tested and localizes to the nucleus, primarily the nucleolus. ZNF346 is exported by exportin-5 but translocates back into nuclei by a facilitated diffusion mechanism. ZNF346 interacts with ILF3 in an RNA-independent manner and may be involved in cell growth and survival.

Description: Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Description: The p53 tumor-suppressor gene integrates numerous signals that control cell life and death; loss of its functions contributes to the development of most cancers. Recent studies have demonstrated the ability of p53 to regulate the expression of several proteins involved in glycolysis and oxidative phosphorylation, such as TIGAR, SCO2, and phosphoglycerate mutase. TIGAR is a recently discovered protein that functions to regulate glycolysis and protect cells against oxidative stress. TIGAR is similar in structure to proteins in the phosphoglycerate mutase family, most notably 6-phosphofructo-2-kinase, suggesting TIGAR may function as a fructose bisphosphatase. Expression of TIGAR in transfected cells correlated with an inhibition of glycolysis and decreased levels of reactive oxygen species and p53-induced apoptosis, indicating that TIGAR may act to modulate the apoptotic response to p53, thereby allowing cells to survive mild or transient stresses.

Description: The nuclear factor NF-kB plays key roles in development and immunity. ABIN3 (A20-binding inhibitor of NF-kB activation 3), also known as TNFAIP3-interacting protein 3 (TNIP3), is a novel negative feedback regulator of LPS-induced NF-kB activation. ABIN3 is a 39 kDa protein that negatively regulates NF-kB activation in response to TNF and LPS. ABIN3 is highly expressed in brain, thymus, lymph node, lung and fetal liver, with low expression in kidney, bone marrow. Through its interaction with A20, ABIN3 interferes with TRAF2-mediated transactivation signals and NF-kB inhibition is mediated by the ABIN-homology domain 2. ABIN3 has been found to be induced by Listeria infection and can be slightly downregulated by dexamethasone. Enhanced expression of ABIN3 in monocytes is associated with sepsis. Thus, ABIN3 is an IL-10-induced gene product capable of attenuating NF-kB in human macrophages yet is inoperative in mice and represents a basis for species-specific differences in IL-10 actions. At least four isoforms of ABIN3 are known to exist.
