Description: The monoclonal antibody DX5 reacts with CD49b, also known as very late antigen-2 (VLA-2) and α2 integrin. The antibody stains the majority of NK cells and a small subpopulation of T cells in all mouse strains tested (e.g., A/J, AKR, BALB/c, C3H/HeJ, C57BL/6, C57BL/10, C57BR, C58, CBA/Ca, CBA/J, DBA/1, DBA/2, SJL, SWR). Three-color flow cytometry analysis has revealed that most cells that express NK-1.1 (NKR-P1C) also express CD49b. However, small subsets of DX5+NK-1.1- and DX5-NK-1.1+ cells can be found, especially among the CD3+ population of T lymphocytes. NK cells cultured in the presence of IL-2 progressively lose reactivity with DX5 as a consequence of cellular proliferation. Therefore, DX5 can be used to define functionally distinct subsets of murine NK cells. The monoclonal antibody DX5 has not been demonstrated to have activating or blocking activity.

Description: CD86, also known as B7-2, is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily of cell surface receptors. It is expressed at high levels on resting peripheral monocytes and dendritic cells and at very low density on resting B and T lymphocytes. CD86 expression is rapidly upregulated by B-cell specific stimuli with peak expression at 18-42 hours after stimulation. CD86, along with CD80 (B7-1), is an important accessory molecule in T cell costimulation via its interaction with CD28 and CD152 (CTLA-4). Since CD86 has rapid kinetics of induction, it is believed to be the major CD28 ligand expressed early in the immune response. The monoclonal antibody GL1 blocks mixed lymphocyte reactions and stimulation of T cells by antigen-presenting cells.

Description: CD90 (Thy-1), a GPI-anchored molecule and one of the smallest members of the immunoglobulin superfamily of cell surface receptors, consists of a single V-set domain. It is expressed on thymocytes, peripheral T lymphocytes, some intraepithelial T lymphocytes, and neurons of all mouse strains. Description: CD43, a member of the sialomucin family of cell surface receptors, is a 95-135 kDa type I transmembrane glycoprotein whose variability in molecular weight is dependent upon the nature of the extracellular O-glycans. It is the major sialoglycoprotein on thymocytes, T cells, and neutrophils. It is also present on activated B cells, plasma cells, NK cells, granulocytes, monocytes, macrophages, platelets, and bone marrow hematopoietic stem cells. CD43, which binds to ICAM-1 (CD54), appears to function as an anti-adhesion molecule, inhibiting T cell interactions including T cell killing, and by increasing the threshold for T cell activation.

Description: The IL-2 receptor (IL-2R) exists in three alternative forms made up from the individual components of CD25, CD122, and CD132. CD25 represents the low affinity α chain of the IL-2R, a type I transmembrane glycoprotein containing two CCP domains. It is rich in O-linked carbohydrates and has a short cytoplasmic tail. CD25 is expressed on activated T cells, B cells, NK cells, and monocytes of all mouse strains tested. It is transiently expressed at a low level during B cell development in bone marrow on the CD45R/B220low TdT- sIg- pre-B II and CD45R/B220low TdT- sIgM+sIgD- immature B cells, but not on the CD45R/B220low TdT+ sIg- pro-B/pre-B I stage nor on CD45R/B220high TdT- sIgM+sIgD+ mature B cells. CD25 is expressed at a higher level during very early T-cell development in fetal and adult thymus. The 7D4 monoclonal antibody reacts with an epitope between amino acids 125 and 212 that is distinct from that recognized by the monoclonal antibody 3C7 (SB Cat. No. 1600). While the monoclonal antibody 7D4 does not block the binding of IL-2 to CD25, when used in combination with the monoclonal antibody 3C7 it results in higher levels of inhibition of IL-2-driven proliferation than either of these two monoclonal antibodies alone.

Description: CD69, also known as very early activation (VEA) antigen, is a disulfide-linked transmembrane homodimer whose differentially glycosylated subunits range from 35-39 kDa. It is a C-type lectin most closely related to the NKR-P1 and Ly-49 NK cell-activation molecules. CD69 is widely expressed on hematopoietic cells, including lymphocytes, neutrophils and eosinophils. Although not detectable on resting lymphocytes, its expression is rapidly upregulated upon activation of T, B and NK cells, and neutrophils. Constitutive expression of CD69 on subsets of thymocytes suggests that it may be involved in regulation of developmental events in addition to its role in activation of a variety of hematopoietic cells. The monoclonal antibody H1.2F3 augments PMA-induced T-cell proliferation and induces redirected lysis of Fc receptor-bearing target cells by NK cells.
